: Glioblastoma multiforme (GBM), a solid tumor that develops from astrocytes, is one of the most aggressive types brain cancer. While there have been improvements in efficacy treating GBM, many problems remain, especially with traditional therapy methods. Therefore, recent studies extensively focused on developing novel therapeutic agents for combating glioblastoma. Natural polyphenols studied their potential as chemopreventive and chemotherapeutic due to wide range positive qualities, including antioxidant, antiinflammatory, cytotoxic, antineoplastic, immunomodulatory activities. Damage to vascular endothelial cells (VECs) and smooth muscle (VSMCs) caused by oxidized low-density lipoprotein (oxLDL) contributes cardiovascular cerebrovascular diseases. Protection effects of Berberine (BBR) on the system have been reported, however, molecular mechanism protection is still unclear. In this study, we established two hyperlipidemia models in zebrafish VEC-VSMC co-culture using high-cholesterol food (HCF) oxLDL, respectively. We demonstrated that HCF doubled total cholesterol glyceride levels, BBR decreased these indices a concentration-dependent manner. Lipid staining hematoxylin-eosin revealed inhibited oxLDL-induced VSMC bulge-like proliferation migration toward VECs prevented HCF-induced trunk obstruction zebrafish. Immunoblot analysis, cell immunofluorescence, co-immunoprecipitation assays, transmission electron microscopy showed oxLDL/HCF increased lectin-like oxLDL receptor-1 (LOX-1) expression at least 5-fold significantly autophagolysosome formation blood vessel These observations were associated with endothelial-to-mesenchymal transition (EMT) triggered VE-cadherin ectopic VSMCs, they responsible for aberrant occlusion.
