Steroidogenic factor-1 (SF-1, also termed Ad4BP; NR5A1 in the official nomenclature) is a nuclear receptor transcription factor that plays crucial role regulation of adrenal and gonadal development, function maintenance. In addition to its classical regulating expression P450 steroid hydroxylases other steroidogenic genes, involvement key processes such as cell survival/proliferation cytoskeleton dynamics have been highlighted for SF-1. SF-1 has restricted pattern expression, being expressed along hypothalamic-pituitary axis organs since time their establishment. Reduced affects proper organogenesis function. On hand, overexpression found adrenocortical carcinoma represents prognostic marker patients’ survival. This review focused on current knowledge about importance dosage gland development function, from cortex formation tumorigenesis. Overall, data converge towards player complex network transcriptional within dosage-dependent manner.

Prostate cancer (PCa) is an age-associated malignancy with high morbidity and mortality rate, posing a severe threat to public health. Cellular senescence, specialized cell cycle arrest form, results in the secretion of various inflammatory mediators. In recent studies, senescence has shown essential role tumorigenesis tumor development, yet extensive effects PCa have not been systematically investigated. Here, we aimed develop feasible senescence-associated prognosis model for early identification appropriate management patients PCa. The RNA sequence clinical information available from Cancer Genome Atlas (TCGA) list experimentally validated senescence-related genes (SRGs) CellAge database were first obtained. Then, senescence-risk signature related was constructed using univariate Cox LASSO regression analysis. We calculated risk score each patient divided them into high-risk low-risk groups terms median value. Furthermore, two datasets (GSE70770 GSE46602) used assess model. A nomogram built by integrating characteristics, which further verified ROC curves calibrations. Finally, compared differences microenvironment (TME) landscape, drug susceptibility, functional enrichment among different groups. established unique prognostic based on eight SRGs, including CENPA, ADCK5, FOXM1, TFAP4, MAPK, LGALS3, BAG3, NOX4, well prognosis-predictive power independent datasets. associated age TNM staging, calibration chart presented consistency prediction. Additionally, could serve as prediction factor due its accuracy. Notably, found that positively mutation burden (TMB) immune checkpoint, whereas negatively correlated dysfunction exclusion (TIDE), suggesting these scores more sensitive immunotherapy. Drug susceptibility analysis revealed responses general drugs (docetaxel, cyclophosphamide, 5-Fluorouracil, cisplatin, paclitaxel, vincristine) yielded between Identifying SRG-score may become promising method predicting tailoring treatment strategies.

Metastatic castration-resistant prostate cancer (mCRPC) is an aggressive form of unresponsive to androgen deprivation therapy (ADT) that spreads quickly other organs. Despite reduced levels after ADT, mCRPC development and lethality continues be conducted by the receptor (AR) axis. The maintenance AR signaling in a result alterations, intratumoral production, action regulatory elements, such as noncoding RNAs (ncRNAs). ncRNAs are key elements signaling, acting tumor growth, metabolic reprogramming, progression. In (PCa), have been reported associated with expression, PCa proliferation, castration resistance. this study, we aimed reconstruct lncRNA-centered network identify lncRNAs which act master regulators (MRs).We used publicly available RNA-sequencing infer mCRPC. Five gene signatures were employed conduct regulator analysis. Inferred MRs then subjected functional enrichment symbolic regression modeling. latter approach was applied greater predictive capacity potential biomarker mCRPC.We identified 31 involved cellular metabolism, invasion-metastasis cascade. Abstract Adrenocortical carcinoma (ACC) is a rare and highly heterogeneous disease with notably poor prognosis due to significant challenges in diagnosis treatment. Emphasizing on the importance of precision medicine, there an increasing need for comprehensive genomic resources alongside well-developed experimental models devise personalized therapeutic strategies. We present ACC_CellMinerCDB, substantive drug sensitivity database (available at https://discover.nci.nih.gov/acc_cellminercdb) comprising ACC cell lines, patient-derived xenografts, surgical samples, responses more than 2,400 drugs examined by NCI National Center Advancing Translational Sciences. This exposes shared pathways among lines thus authenticating as research models. It also allows exploration pertinent treatment markers such MDR-1, SOAT1, MGMT, MMR, SLFN11 introduces potential repurpose agents like temozolomide therapy. ACC_CellMinerCDB provides foundation exploring larger preclinical Significance: responses, reveals treatment-relevant ACC. resource offers insights into targets opportunity existing
