Salivary Gland Cancers in the Era of Molecular Analysis: The Role of Tissue and Liquid Biomarkers

Background:
Salivary
gland
cancers
(SGCs)
are
a
rare
and
heterogeneous
group
of
malignancies,
accounting
for
approximately
5%
head
neck
cancers.
Despite
their
rarity,
advances
in
molecular
profiling
have
revealed
variety
genetic
pathways,
many
which
potentially
actionable
with
targeted
therapies.
Methods:
We
reviewed
the
current
literature
involving
landscape
SGCs,
encompassing
diagnostic
prognostic
value
tissue
liquid
biomarkers
potential
therapeutic
targets
across
various
histological
subtypes.
Results:
Our
review
highlights
key
findings
such
as
CRTC1-MAML2
fusion
mucoepidermoid
carcinoma
MYB-NFIB
rearrangements
adenoid
cystic
carcinoma,
but
also
targetable
alterations
HER2
AR
positivity
salivary
duct
ETV6-NTRK3
secretory
carcinoma.
Liquid
biopsy
(both
blood-
or
salivary-based),
including
circulating
tumor
DNA,
cells,
miRNAs,
offers
novel,
noninvasive
approaches
disease
monitoring
personalized
treatment.
Emerging
therapies
inhibitors,
androgen
deprivation
therapy,
TRK
inhibitors
underscore
shift
towards
precision
oncology
managing
these
malignancies.
Conclusions:
promising
advances,
challenges
remain
due
to
rarity
phenotypic
heterogeneity
emphasizing
need
molecularly
stratified
clinical
trials.
This
presents
an
overview
biomarkers,
focusing
on
innovations
that
lay
foundation
improved
treatment
strategies
SGCs.

plays
a
multifaceted
role
in
oral
health
and
systemic
well-being.
It
supports
digestion,
protects
tissues,
maintains
healthy
microbiome,
facilitates
wound
healing.
Additionally,
saliva
serves
as
diagnostic
tool
that
reflects
disease/therapeutic
states.
Furthermore,
although
shows
protective
effect
against
cancer
development,
once
tumor
formation
occurs,
it
may
be
involved
progression
metastasis
via
exosomes
microRNAs.
This
review
discusses
the
essential
of
saliva;
its
relationship
with
progression,
head
neck
squamous
cell
carcinoma
(HNSCC);
liquid
biopsy
tools
for
early
diagnosis
monitoring
HNSCC;
potential
therapeutic
agents.
