ABSTRACT Uterine fibroids (UFs) are the most common benign gynecologic tumours affecting women of reproductive age. This study aims to deepen understanding UFs complex aetiology through harnessing power 3D organoid models derived from human myometrial stem cells emulate in vivo behaviour these tumours. Isolated SCs were cultured over 7 days under a defined culture system. Immunohistochemistry, Immunofluorescence, stiffness, RNA Sequencing was conducted, and differential gene expression assessed using RT‐PCR. Uterine leiomyoma is one of the most common gynaecological diseases, which often leads to loss fertility. The aim study was determine levels melatonin, reproductive hormones state angiogenesis, antioxidant system lipid peroxidation women age who were diagnosed with uterine leiomyoma. Material methods. 60 (study group) examined. control group consisted 20 healthy same group. Melatonin women's blood determined once, on an empty stomach, at 8 a.m.; concentrations estradiol progesterone follicular phase menstrual cycle also measured. angiogenesis studied examining vascular endothelial growth factor (VEGF) content final metabolites nitric oxide NO plasma. activity assessed plasma reduced glutathione, glutathione peroxidase, transferase. Indicators investigated malondialdehyde (MDA) erythrocytes. Abstract The role of long non‐coding RNAs in fibroid pathogenesis remains largely unexplored. In a previous study, we found elevated XIST (X‐inactive specific transcript) levels fibroids, which sponged miR‐29c and miR‐200c, leading to the overexpression their target genes. This study aimed assess therapeutic potential downregulation treatment. Ovariectomized SCID (severe combined immunodeficiency) mice were implanted with tumors transduced siRNA or control via lentivirus.
