Zinc finger proteins (ZFP) are metalloproteins whose zinc atom interacts with side chains of specific histidines and cysteines the motif, generating functional three dimensional structures. Background Rapidly developed chemoresistance to dacarbazine (DTIC) is a major obstacle in the clinical management of melanoma; however, roles and mechanisms epi-transcriptomic RNA modification this process have not been investigated. Method DTIC-resistant (DR) melanoma cells were established for bulk sequencing. The expressions mRNAs detected using qRT-PCR, protein levels determined Western blotting immunohistochemistry. Acetylated RNAs by dot immunoprecipitation sequencing (acRIP-seq). A lung metastasis mouse model was evaluate anti-melanoma effects vivo . Results We identified that expression N -acetyltransferase 10 (NAT10), catalytic enzyme 4 -acetylcytidine (ac4C) RNA, significantly upregulated DR cells. Clinically, NAT10 elevated disease progression samples predicted poor outcome. Using ac4C (ac4C-RIP), we found two C2H2 zinc finger transcriptional factors, DDX41 ZNF746 , targets NAT10-mediated modification.
