Melanogenesis is a biochemical process that regulates skin pigmentation, which crucial role in protecting against ultraviolet radiation. It also associated with hyperpigmentation conditions such as melasma and age spots, negatively impact aesthetics self-confidence. Tyrosinase (TYR), key enzyme the melanogenesis pathway, catalyzes biosynthesis of melanin skin. Inhibition tyrosinase particularly by blocking its active site preventing binding natural substrates tyrosine, can reduce production, making it promising therapeutic target for treating hyperpigmentation. Peptides have emerged therapeutics to regulate minimizing side effects conventional whitening therapeutics. Malignant melanoma (MM) is a highly aggressive cancer. Different subtypes have different sensitivities to immunotherapy and lack peripheral blood markers. Few studies examined the role of inflammatory markers in predicting overall survival (OS) stage IV acral (AM) cutaneous (CM) patients receiving immunotherapy. This study aimed investigate value efficacy prognosis for who underwent multicenter reviewed clinicopathological characteristics 94 AM CM PD-1 inhibitors therapy. Pearson's chi-squared test or Fisher's exact was used compare baseline characteristics. Abstract Objectives Lipid metabolic dysregulation plays a critical role in the biological behavior of skin cutaneous melanoma (SKCM). Hence, we aimed to identify lipid metabolism-related genes (LMGs) and possible prognostic models for SKCM, perform risk assessment, predict effective therapies. Methods SKCM patient data were downloaded from The Cancer Genome Atlas (TCGA) used as training set; GSE65904 was validation dataset. A model established by multivariate Cox regression analysis LASSO algorithm. samples sets grouped into high- low-risk groups, respectively, accordance with model, score (RS) distribution survival ROC curve obtained. ‘limma’ package R3.6.1 Version 3.34.7 filter significant differentially expressed (DEGs) set between groups.
