The dapsone/flavone cocrystal system served as a benchmark for both experimental and virtual screening methods. Expanding beyond this, two additional active pharmaceutical ingredients (APIs), sulfanilamide sulfaguanidine, structurally related to dapsone were chosen investigate the impact of substituents on formation. involved mechanochemical methods, slurry experiments, hot-melt extrusion, contact preparation method. focused crystal structure prediction (CSP), molecular complementarity, hydrogen-bond propensity, electrostatic potentials. CSP studies not only indicated that each three APIs should form cocrystals with flavone but also reproduced known single- multicomponent phases. Experimentally, ACC, BCC, CCC, DCC reproduced, CCC was identified nonstoichiometric hydrate, fifth (ECC), t-butanol solvate, discovered. polymorphs ACC BCC are enantiotripically related, DCC, exhibiting different stoichiometric ratio, is enthalpically stabilized over other cocrystals. For sulfaguanidine/flavone system, novel, identified. structures polymorph solved from powder X-ray diffraction data, stability all assessed through differential scanning calorimetry lattice energy calculations. Despite computational indications, diverse array cocrystallization techniques did result in sulfanilamide/flavone cocrystal. driving force behind dapsone's tendency cocrystallize can be attributed overall strength interactions potential strong API···API API···coformer contributing factor.
