Dietary modifications to overcome infertility have attracted attention; however, scientifically substantiated information on specific dietary components affecting fertility and their mechanisms is limited. Herein, we investigated diet-induced, reversible in female mice lacking the heterodimer of ATP-binding cassette transporters G5 G8 (ABCG5/G8), which functions as a lipid exporter intestine. We found that phytosterols, especially β-sitosterol brassicasterol, are substrates ABCG5/G8, potent but reproductive toxicities mice. Mechanistically, these phytosterols inhibited ovarian folliculogenesis reduced egg quality by enhancing polycomb repressive complex 2-mediated histone H3 trimethylation at lysine 27 ovary. Clinical analyses showed serum phytosterol levels were significantly negatively correlated with blastocyst development rate fertilized eggs women undergoing vitro fertilization, suggesting affect both humans Thus, avoiding excessive intake certain would be beneficial for health. In mouse ovary, shown inhibit reduce PRC2-mediated trimethylation.
