Intergenerational trauma, violence, and maltreatment, in which symptoms or experiences of an ancestor's trauma repeat otherwise manifest subsequent generations, presents a weighty societal challenge to multiplicity therapeutic intervention strategies have been applied. Theoretical perspectives are antecedent clinical social intervention, informing decisions both policy practice. However, these frequently remain subliminal imperceptible the discourse, resulting interventions that somewhat dislocated from their theoretical foundations. This narrative review seeks summarize discuss each theories as they apply intergenerational reveal potential association with specific models approaches. It positions flexibility between integration opportunities reach new enhanced understandings engender distinctive interventions. An enriched understanding explaining deeper appreciation for pertinence theory practice, incitement blend unique ways is, herewith, reached.

Early-life adversities, whether prenatal or postnatal exposure, have been linked to adverse mental health outcomes later in life increasing the risk of several psychiatric disorders. Research on its neurobiological consequences demonstrated an association between exposure adversities and persistent alterations structure, function, connectivity brain. Consistent evidence supports idea that regulation gene expression through epigenetic mechanisms are involved embedding impact early-life experiences genome mediate social environments behavioral phenotypes. In addition, studies from rodent models humans suggest these acquired factors can be transmitted offspring following generations potentially contributing a cycle disease risk. However, one important aspects mechanisms, unlike genetic sequences fixed unchangeable, is although markings long-lasting, they nevertheless reversible. this review, we summarize our current understanding derived malnutrition, maltreatment poverty, with huge pervasive health. We also discuss about transgenerational inheritance mammals experimental data suggesting suitable pharmacological interventions could reverse modifications induced by negative experiences. regard, must accompanied efforts determine causes promote result inequity population.

Stress, encompassing psychological, physical, and physiological challenges, is an important factor affecting individual's well-being potentially leading to psychiatric, neurodegenerative, immune, metabolic disorders. However, not everyone exposed stress develops these conditions, highlighting the concept of resilience. Resilience a dynamic process categorized into four dimensions: pre-existing resilience capacity, ongoing processes, post-stress outcomes, recovery from psychopathologies. These dimensions involve genomic, cellular, systemic interactions influenced by genetic factors, early life experiences, adult experiences in addition community/environmental health behaviors. The biological response encompasses endocrine, autonomic, immunological, behavioral components, modulated stressor characteristics individual traits. Due limitations studying humans, translational models using rodents cell cultures are essential. Rodent include acute, chronic, traumatic paradigms, aiding study stress-related molecular outcomes. Additionally, models, such as prenatal maternal separation, provide insights developmental impacts. In this review, first, rodent for lifelong exposure will be summarized considering their validity, advantages, limitations. Subsequently, overview designed enhance capacity rodents, later employed outcomes given. Lastly, focus shifted culture iPSCs models. Finally, future considerations focused on improving used discussed. It aimed designs access more effective biomarkers associated with Stress complex phenomena various spanning levels. Integrating data across remains crucial unraveling complexities disorders

The early postnatal period is a critical neurodevelopmental stage characterized by rapid neural maturation and adversely affected early-life stressors. This study explored the behavioural, physiological, epigenetic consequences of stress in population homeless rescue kittens. longitudinal included 50 kittens rescued placed into foster care Prince Edward Island Humane Society. They underwent behavioural testing at 8, 10, 12 weeks age. Hair cortisol concentration was measured 8 served as physiological marker previous 3 months’ cumulative response, which, for these kittens, late gestation period. A blood sample relative telomere length measurement taken 10–12 to estimate changes young Data were analyzed with respect age performance all repeated measures tests, status stray or surrender, presence dam their homes. As expected, tests changed over 5 testing. Kittens separated from mothers exhibited significantly higher hair concentrations (p = 0.02) elongated lengths 0.04). No correlation found between 0.99). These results support need further on effects epigenetics stress, both across species.

Abstract Background Posttraumatic stress disorder (PTSD) affects approximately 8% of the US population, with varying susceptibility among individuals exposed to trauma. While genetic factors contribute PTSD risk, emerging evidence suggests that epigenetic mechanisms play a crucial role in translating environmental exposures into lasting neurobiological changes. Purpose This review provides comprehensive analysis cutting-edge research on PTSD, particular emphasis novel findings regarding resilience and mechanisms. We explore recent technological advances their applications understanding pathophysiology. Main body Advanced epigenomic approaches have revealed complex interactions between DNA methylation, histone modifications, non-coding RNAs PTSD. Novel highlight cell type-specific signatures temporal dynamics following trauma exposure. Single-cell studies identified previously unknown cellular heterogeneity responses. Recent data modifications not only influence individual but may also transgenerational transmission effects. Integrative multi-omics new insights molecular networks underlying vulnerability. Conclusion unprecedented complexity These open avenues for personalized interventions based profiles suggest therapeutic strategies targeting modifications. enhanced has significant implications risk assessment, prevention, treatment. Abstract Stress in early life can affect the progeny and increase risk to develop psychiatric cardiometabolic diseases across generations. The cross-generational effects of stress have been modeled mice demonstrated be associated with epigenetic factors germline. While is known gut microbial features, whether its persist passed not well defined. Here we show that postnatal shifts fecal composition (binary Jaccard index) throughout life, including abundance eight amplicon sequencing variants (ASVs). Further on composition, structure (weighted index), 16 ASVs are detected two These accompanied by changes bacterial metabolites any generation. results suggest community induced traumatic perpetuated from exposed parent offspring.
