Abstract: Over the past decade, idea of targeting endocannabinoid system to treat anxiety disorders has received increasing attention. Previous studies focused more on developing cannabinoid receptor agonists or supplementing exogenous cannabinoids, which are prone various adverse effects due their strong pharmacological activity and poor selectivity, limiting application in clinical research. Endocannabinoid hydrolase inhibitors considered be most promising development strategies for treatment disorders. More recent efforts have emphasized that inhibition two major endogenous hydrolases, monoacylglycerol lipase (MAGL) fatty acid amide (FAAH), indirectly activates receptors by levels synaptic gap, circumventing desensitization resulting from direct enhancement signaling. Traumatic muscle injuries (TMIs) and pain (MP) negatively impact athletes’ performance quality of life. Both conditions have a complex pathophysiology involving the interplay between genetic environmental factors. Yet, existing data are scarce controversial. To provide more insights, this study aimed to investigate association single-nucleotide polymorphisms (SNPs) previously linked athletic status with TMI MP after exercise among Brazilian high-performance athletes from different sports modalities (N = 345). The important determinants was also assessed. From six evaluated SNPs (ACTN3 rs1815739, FAAH rs324420, PPARGC1A rs8192678, ADRB2 rs1042713, NOS3 rs1799983, VDR rs731236), none significantly associated TMI. Regarding exercise, ACTN3 rs1815739 (CC/CT vs. TT; adjusted odds ratio (aOR) 1.90; 95% confidence interval (95%Cl), 1.01–3.57) rs324420 (AA AC/CC; aOR 2.30; 95%Cl, 1.08–4.91) were independent predictors according multivariate binomial analyses for age (≥23 <23 years), sex (male female), tobacco consumption (yes no).
