Abstract Obesity is a complex, chronic, and multifactorial condition characterized by abnormal fat accumulation in tissues organs inducing negative effects on human health. Alzheimer's disease progressive irreversible neurodegenerative disease, associated with amyloid plaques neurofibrillary tangles the brain. The correlation between obesity has been discovered, but molecules molecular mechanisms linking these conditions are not yet fully elucidated. In this review, we focused most important processes alterations of brain structure functions.

This study is the first to evaluate oxidative stress biomarkers in saliva/blood of patients with varying degrees dementia progression. The included 50 healthy controls and divided into two groups: those mild moderate (MMSE 11-23) suffering from severe 0-10). Cognitive functions subjects were assessed using Mini Mental State Examination (MMSE). Enzymatic non-enzymatic antioxidants, damage products protein glycoxidative modifications determined non-stimulated (NWS) stimulated (SWS) saliva as well erythrocyte/plasma samples. Generally, patients, we observed depletion antioxidant defences leading NWS, SWS blood Both salivary increased severity disease, correlated a decrease cognitive functions. Interestingly, reduced glutathione (GSH) NWS not only dementia, but also GSH concentration plasma. In receiver operating characteristic (ROC) analysis, have demonstrated that clearly distinguishes or (area under curve (AUC) = 1). Therefore, can be used non-invasive biomarker impairment.

Down syndrome (DS) is the most frequent chromosomal abnormality that causes intellectual disability, resulting from presence of an extra complete or segment chromosome 21 (HSA21). In addition, trisomy HSA21 contributes to altered energy metabolism appears be a strong determinant in development pathological phenotypes associated with DS. Alterations include mitochondrial defects, increased oxidative stress levels, impaired glucose and lipid among others, finally reduced production cellular dysfunctions. These molecular defects seem account for high incidence metabolic disorders, i.e., diabetes and/or obesity as well higher risk develop Alzheimer disease (AD) A dysregulation insulin signaling downstream pathways represents common pathophysiological aspect both peripheral central alterations leading diabetes/obesity AD. This further strengthened by evidence showing mechanisms responsible such appear similar between organs brain. Considering DS subjects are at either brain this review will discuss current knowledge about link insulin-related Drawing signature underlying these processes key challenge identify novel drug targets set-up new prevention strategies aimed reduce impact disorders cognitive decline.
