Age-related impairment of the diaphragm causes respiratory complications. Neuromuscular junction (NMJ) dysfunction can be one triggering events in weaknesses old age. Prominent structural and functional alterations NMJs were described elderly rodents, but NMJ changes middle age remain unclear. Here, we compared muscles from young adult (3 months) middle-aged (12 BALB/c mice. Microelectrode recordings, immunofluorescent staining, electron microscopy, myography, whole-body plethysmography used. We revealed presynaptic (i) postsynaptic (ii) changes. The former included an increase both action potential propagation velocity neurotransmitter release evoked by low-, moderate-, high-frequency activity a decrease immunoexpression synapsin 1 synaptic vesicle clustering. latter consisted currents via nicotinic acetylcholine receptors area their distribution. It is known that plantar mechanical stimulation (PMS) able to attenuate unloading-induced skeletal muscle atrophy and impaired function. However, molecular mechanisms underlying the effect of PMS on during unloading remain undefined. The aim study was evaluate effects anabolic catabolic signaling pathways in rat soleus at early stages unloading. Wistar rats were randomly assigned ambulatory control, hindlimb suspension (HS) for 1 or 3 days, HS days with PMS. key markers assessed by western blotting RT-PCR. Protein synthesis (PS) rate estimated using SUnSET technique. attenuated a 1-day HS-induced decrease 4E-BP1, GSK-3β, AMPK phosphorylation. also partially prevented PS, phosphorylation nNOS an increase eEF2 after 3-day HS. 1-and MuRF-1, but not MAFbx, upregulation did affect ribosome biogenesis (18S+28S rRNA, c-myc) as well AKT Thus, global PS which accompanied attenuation MuRF-1 mRNA expression changes

Neuromuscular disorders (NMDs) affect 1 in 3000 people worldwide. There are more than 150 different types of NMDs, where the common feature is loss muscle strength. These classified according to their neuroanatomical location, as motor neuron diseases, peripheral nerve neuromuscular junction and diseases. Over years, numerous studies have pointed protein homeostasis a crucial factor development these fatal The ubiquitin–proteasome system (UPS) plays fundamental role maintaining homeostasis, being involved degradation, among other cellular functions. Through cascade enzymatic reactions, proteins ubiquitinated, tagged, translocated proteasome be degraded. Within ubiquitin system, we can find three main groups enzymes: E1 (ubiquitin-activating enzymes), E2 (ubiquitin-conjugating E3 (ubiquitin–protein ligases). Only ubiquitinated with specific chain linkages (such K48) will degraded by UPS. In this review, describe relevance summarizing UPS that been pathological conditions disorders, such Spinal Muscular Atrophy (SMA), Charcot–Marie–Tooth disease (CMT), or Duchenne Dystrophy (DMD), others. A better knowledge processes maintenance proteostasis may pave way for future progress disorder treatments.
