With the progression sequencing methods modern bioinformatics tools, novel roles microRNAs mechanisms pathophysiological states continue to expand. Advances detection have further enabled larger adoption studies utilizing minimal sample volumes, allowing analysis low-volume biofluids, aqueous humor tear fluid. The reported abundance extracellular these biofluids has prompted explore their biomarker potential. This review compiles current literature reporting human fluid association with ocular diseases including dry eye disease, Sjögren’s syndrome, keratitis, vernal keratoconjunctivitis, glaucoma, diabetic macular edema, retinopathy, well non-ocular diseases, Alzheimer’s breast cancer. We also summarize known shed light on future this field.

Heterogeneity of triple-negative breast cancer is well known at clinical, histopathological, and molecular levels. Genomic instability greater mutation rates, which may result in the creation neoantigens enhanced immunogenicity, are additional characteristics this type. Clinical outcome poor due to early age onset, high metastatic potential, increased likelihood distant recurrence. Consequently, efforts elucidate mechanisms development, progression, spread have been initiated improve treatment options outcomes for these patients. The extremely complex heterogeneous tumor immune microenvironment made up several cell types commonly possesses disorganized gene expression. Altered signaling pathways mainly associated with mutated genes including p53, PIK3CA, MAPK, positively correlated regulating response. Of note, particular immunity-associated could be used prognostic indexes assess most effective management. Recent findings highlight fact that long non-coding RNAs also play an important role shaping formation, can mediate evasion. Identification signatures, through use multi-omics approaches, effector drive stages carcinogenic process steps developing new strategies targeted prevention. Advances immunotherapy by remodeling host system eradicate cells great promise lead novel therapeutic strategies. Current research focused on combining checkpoint inhibition chemotherapy, PARP inhibitors, vaccines, or natural killer therapy. Targeted therapies response, eliminate resistance, overall patient survival. In future, evolving advancements should implemented personalized medicine state-of-art management

In recent years, the importance of epigenetic markers in carcinogenesis different malignant neoplasms has been demonstrated, also demonstrating their utility for understanding metastatic spread and tumor progression cancer patients. Among biomarkers, microRNAs represent a set non-coding RNAs that regulate gene expression, having involved wide variety neoplasia acting oncogenic pathways. Both overexpression downregulation complex interaction with various genes whose ultimate consequence is increased cell proliferation, invasion driver markers. It should be noted current clinical practice, even though combination shown to useful by authors at diagnostic prognostic levels, there are no kits can used initial approach or assess recurrences oncological diseases. Previous works have cited as critical role several carcinogenic mechanisms, ranging from cycle alterations angiogenesis mechanisms distant dissemination. Indeed, specific seem tightly modulation components related these processes. For instance, cyclins cyclin-dependent kinases, transcription factors, signaling molecules angiogenic/antiangiogenic products, among others, recognized targets types cancer. Therefore, purpose this article describe main implications alterations, metastasis angiogenesis, trying summarize involvement carcinogenesis.

Breast cancer (BC) is characterized by an extensive genotypic and phenotypic heterogeneity. In-depth investigations into the molecular bases of BC phenotypes, carcinogenesis, progression, metastasis are necessary for accurate diagnoses, prognoses, therapy assessments in predictive, precision, personalized oncology. This review discusses both classic as well several novel omics fields that involved or should be used modern investigations, which may integrated a holistic term, onco-breastomics. Rapid recent advances profiling strategies analytical techniques based on high-throughput sequencing mass spectrometry (MS) development have generated large-scale multi-omics datasets, mainly emerging from three ”big omics”, central dogma biology: genomics, transcriptomics, proteomics. Metabolomics-based approaches also reflect dynamic response cells to genetic modifications. Interactomics promotes view research constructing characterizing protein–protein interaction (PPI) networks provide hypothesis pathophysiological processes progression subtyping. The emergence new omics- epiomics-based multidimensional opportunities gain insights heterogeneity its underlying mechanisms. main epiomics (epigenomics, epitranscriptomics, epiproteomics) focused epigenetic DNA changes, RNAs modifications, posttranslational modifications (PTMs) affecting protein functions in-depth understanding cell proliferation, migration, invasion. Novel fields, such epichaperomics epimetabolomics, could investigate interactome induced stressors PPI metabolites, drivers BC-causing phenotypes. Over last years, proteomics-derived omics, matrisomics, exosomics, secretomics, kinomics, phosphoproteomics, immunomics, provided valuable data deep dysregulated pathways their tumor microenvironment (TME) immune (TIMW). Most these datasets still assessed individually using distinct approches do not generate desired expected global-integrative knowledge with applications clinical diagnostics. However, hyphenated approaches, proteo-genomics, proteo-transcriptomics, phosphoproteomics-exosomics useful identification putative biomarkers therapeutic targets. To develop non-invasive diagnostic tests discover BC, omics-based allow significant blood/plasma-based omics. Salivaomics, urinomics, milkomics appear integrative high potential early diagnoses BC. Thus, analysis circulome considered frontier liquid biopsy. Omics-based modeling, classification subtype characterization. future single-cell analyses.

Growing evidence suggests that, among the different molecular/cellular pathophysiological mechanisms associated with cancer, there are 14 hallmarks that play a major role, including: (i) sustaining proliferative signaling, (ii) evading growth suppressors, (iii) activating invasion and metastasis, (iv) enabling replicative immortality, (v) inducing angiogenesis, (vi) resisting cell death, (vii) reprogramming energy metabolism, (viii) immune destruction, (ix) genome instability mutations, (x) tumor-promoting inflammation, (xi) unlocking phenotypic plasticity, (xii) nonmutational epigenetic reprogramming, (xiii) polymorphic microbiomes, (xiv) senescent cells. These also development of breast which represents most prevalent tumor type in world. The present narrative review aims to describe, for first time, effects physical activity/exercise on these hallmarks. In summary, an active lifestyle, particularly regular exercise, provides beneficial all might therefore help counteract progression disease or its burden.
