Brain tumors lead to modifications of brain networks. Graph theory plays an important role in clarifying the principles connectivity. Our objective was investigate network related tumor grade and location using resting-state functional magnetic resonance imaging (fMRI) graph theory. We retrospectively studied 30 low-grade (LGG), high-grade (HGG) left-hemispheric glioma patients 20 healthy controls (HC) with rs-fMRI. Tumor labeled as: frontal, temporal, parietal, insular or occipital. collected patients’ clinical data from records. analyzed whole-brain hemispheric networks all HC. Subsequently, we lobar subgroups divided by location. Seven graph-theoretical metrics were calculated (FDR p < 0.05). Connectograms computed for significant nodes. The two-tailed Student t-test Mann−Whitney U-test (p 0.05) used compare data. analysis showed increased ipsilateral connectivity LGG (global efficiency = 0.03) decreased contralateral HGG (degree/cost 0.028). Frontal temporal bilateral modifications; parietal only local effects. Temporal led a decrease metrics. influence pattern reorganization. may show more favorable changes than HGG, reflecting fewer deficits.

Abstract The current understanding of lactate extends from its origins as a byproduct glycolysis to role in tumor metabolism, identified by studies on the Warburg effect. shuttle hypothesis suggests that plays an important bridging signaling molecule coordinates among different cells, organs and tissues. Lactylation is posttranslational modification initially reported Professor Yingming Zhao’s research group 2019. Subsequent confirmed lactylation vital component function involved proliferation, neural excitation, inflammation other biological processes. An indispensable substance for various physiological cellular functions, regulatory aspects energy metabolism signal transduction. Therefore, comprehensive review summary presented clarify disease provide reference direction future research. This offers systematic overview homeostasis roles pathological processes, well effects diseases, particularly cancer.

Abstract Glioblastoma multiforme (GBM) is the most aggressive and common malignant primary brain tumor. Patients with GBM often have poor prognoses, a median survival of ∼15 months. Enhanced understanding molecular biology central nervous system tumors has led to modifications in their classifications, recent which classified these into new categories made some changes nomenclature grading system. This review aims give panoramic view last 3 years’ findings glioblastoma characterization, its heterogeneity, current advances treatment. Several parameters been used achieve an accurate personalized characterization patients, including epigenetic, genetic, transcriptomic metabolic features, as well age‐ sex‐related patterns involvement several noncoding RNAs progression. Astrocyte‐like neural stem cells outer radial glial‐like from subventricular zone proposed agents involved IDH‐wildtype origin, but this remains controversial. metabolism characterized by upregulation PI3K/Akt/mTOR signaling pathway, promotion glycolytic flux, maintenance lipid storage, other features. also contributes glioblastoma's resistance conventional therapies. Glioblastoma is the most aggressive cancer among primary brain tumours. As with other cancers, incidence of glioblastoma increasing; despite modern therapies, overall mean survival patients post-diagnosis averages around 16 months, a figure that has not changed in many years. Cannabigerol (CBG) only recently been reported to prevent progression certain carcinomas and yet studied glioblastoma. Here, we have compared cytotoxic, apoptotic, anti-invasive effects purified natural cannabinoid CBG together CBD THC on established differentiated tumour cells stem cells. reduced viability both types similar extent, whereas combining was more efficient than THC. CBG, alone combination, induced caspase-dependent cell apoptosis, there no additive effect. Of note, inhibited invasion manner chemotherapeutic temozolomide. We demonstrated little added value combined-cannabinoid treatment, suggesting this psychotropic should be replaced future clinical studies therapy.

The biology and physics underlying glioblastoma is not yet completely understood, resulting in the limited efficacy of current clinical therapy. Recent studies have indicated importance mechanical stress on development malignancy cancer. Various types activate adaptive tumor cell responses that include alterations extracellular matrix (ECM) which an impact malignancy. In this review, we describe discuss knowledge effects ECM GBM aggressiveness. Gradual changes brain been connected to biological physical cells. For example, increased expression several components such as glycosaminoglycans (GAGs), hyaluronic acid (HA), proteoglycans fibrous proteins result stiffening ECM, alters inter- intracellular signaling activity. Several mechanosensing pathways identified orchestrate responses, Hippo/YAP, CD44, actin skeleton signaling, remodel cytoskeleton affect cellular properties cell-cell/ECM interactions, growth, migration/invasion vitro, hydrogels are used a model mimic reconstruct its mechanics, also discuss. Glioblastoma is a highly aggressive and invasive tumor that affects the central nervous system (CNS). With five-year survival rate of only 6.9% median time eight months, it has lowest among CNS tumors. Its treatment consists surgical resection, subsequent fractionated radiotherapy concomitant adjuvant chemotherapy with temozolomide. Despite implementation clinical interventions, recurrence common occurrence, over 80% cases arising at edge resection cavity few months after treatment. The high location glioblastoma indicate need for better understanding peritumor brain zone (PBZ). In this review, we first describe main radiological, cellular, molecular biomechanical tissue features PBZ; subsequently, discuss its current management, potential local therapeutic approaches future prospects.

Betulinic acid is a naturally occurring compound that can be obtained through methanolic or ethanolic extraction from plant sources, as well chemical synthesis microbial biotransformation. has been investigated for its potential therapeutic properties, and exhibits anti-inflammatory, antiviral, antimalarial, antioxidant activities. Notably, ability to cross the blood-brain barrier addresses significant challenge in treating neurological pathologies. This review aims compile information about impact of betulinic an antitumor agent, particularly context glioblastoma. Importantly, demonstrates selective activity against glioblastoma cells by inhibiting proliferation inducing apoptosis, consistent with observations other cancer types. Compelling evidence published highlights acid's action suppressing Akt/NFκB-p65 signaling cascade enhancing cytotoxic effects chemotherapeutic agent temozolomide. Interesting findings also suggest focus on researching reduction glioblastoma's invasiveness aggressiveness profile. involves modulation extracellular matrix components, remodeling cytoskeleton, secretion proteolytic proteins. Glioma is one of the most common primary central nervous system (CNS) tumors, and its molecular diagnosis crucial. However, surgical resection or biopsy risky when tumor located deep in brain brainstem. In such cases, a minimally invasive approach to liquid beneficial. Cell-free DNA (cfDNA), which directly reflects tumor-specific genetic changes, has attracted attention as target for biopsy, blood-based cfDNA monitoring been demonstrated other extra-cranial cancers. it still challenging fully detect CNS tumors derived from blood, including gliomas, because unique structure blood–brain barrier. Alternatively, cerebrospinal fluid (CSF) an ideal source expected contribute significantly gliomas. Several successful studies have conducted alterations CSF using digital PCR and/or next-generation sequencing. This review summarizes current status CSF-based cfDNA-targeted It highlights how approaches differ biopsies cancers discusses issues prospects.
