Cannabis sativa L. contains numerous compounds with antioxidant and anti-inflammatory properties, including the flavonoids cannabinoids, particularly Δ-9-tetrahydrocannabinol (THC) cannabidiol (CBD). Cannabinoids have an effect on endocannabinoid system (ECS), a cellular communication network, are, hence, widely studied for medical applications. Epidiolex®, 99% pure oral CBD extract, has been approved by FDA treatment of epilepsy. Nabiximols (Sativex) is oromucosal spray containing equal volume THC CBD, it commonly used as add-on unresponsive spasticity in multiple sclerosis (MS) patients. Several vitro vivo studies also shown that cannabinoids can be to treat various types cancer, such melanoma brain glioblastoma; first positive clinical trials anticancer THC:CBD blend temozolomide (TMZ) highly invasive cancer are very promising. Abstract The use of cannabidiol (CBD) for therapeutic purposes is receiving considerable attention, with speculation that CBD can be useful in a wide range conditions. Only one product, purified form plant-derived solution (Epidiolex), approved the treatment seizures patients Lennox-Gastaut syndrome, Dravet or tuberous sclerosis complex. Appraisal evidence base complicated by fact products sometimes have additional phytochemicals (like tetrahydrocannabinol (THC)) present, which make identification active pharmaceutical ingredient (API) positive studies difficult. aim present review to critically clinical using only, order establish upcoming indications might beneficial. areas there most support are anxiety (positive data 7 uncontrolled and 17 randomised controlled trials (RCTs)), psychosis schizophrenia 1 study 8 RCTs), PTSD 2 4 RCTs) substance abuse 3 RCTs). Seven improve sleep quality, but this has only been verified small RCT. Limited supports Parkinson’s (3 autism smoking cessation (2 graft-versus-host disease intestinal permeability (1 RCT each). Cannabis is the most used drug of abuse worldwide. It well established that abundant phytocannabinoids in this plant are Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD). These two compounds have remarkably similar chemical structures yet vastly different effects brain. By binding to same receptors, THC psychoactive, while CBD has anxiolytic antipsychotic properties. Lately, a variety hemp-based products, including THC, become widely available food health industry, medical recreational use cannabis been legalized many states/countries. As result, people, youths, consuming because it considered “safe”. An extensive literature exists evaluating harmful both adults adolescents, but little known about long-term exposure, especially adolescence. Evidence suggests that the phytocannabinoids Δ-9-tetrahydrocannabinol (THC) and cannabidiol (CBD) differentially regulate salience attribution psychiatric risk. The ventral hippocampus (vHipp) relays emotional via control of dopamine (DA) neuronal activity states, which are dysregulated in psychosis schizophrenia. Using vivo electrophysiology male Sprague Dawley rats, we demonstrate intra-vHipp THC strongly increases tegmental area (VTA) DA frequency bursting rates, decreases GABA frequency, amplifies VTA beta, gamma ε oscillatory magnitudes modulation local extracellular signal-regulated kinase phosphorylation (pERK1–2). Remarkably, whereas also potentiates morphine place-preference fear conditioning assays, CBD coadministration reverses these changes by downregulating pERK1–2 signaling, as pharmacological reactivation blocked inhibitory properties CBD. These results identify vHipp signaling a critical neural nexus point mediating THC-induced affective disturbances suggest potential mechanism may counteract psychotomimetic psychotropic side effects THC. SIGNIFICANCE STATEMENT Strains marijuana with high levels delta-9-tetrahydrocannabinol low have been shown to underlie neuropsychiatric risks associated high-potency cannabis use. However, mechanisms mitigates not identified. We induces cognitive abnormalities resembling symptoms directly hippocampus, while dysregulating states amplifying frequencies (ERK) pathway. In contrast, ERK phosphorylation, prevented behavioral abnormalities. findings novel molecular account for how functionally

Epidemiological and clinical evidence suggests that diabetes increases the risk of liver cancer. Although co-occurrence type 2 (T2D) cancer is becoming more frequent, underlying mechanisms remain unclear. Even though baicalin, extensively used in traditional Chinese medicine (TCM), can control T2D inhibit separately, minimal research available regarding its possible effect on T2D-induced Thus, present study, we aimed to investigate role baicalin hepatocellular cancer, for first time, particularly emphasized regulation genes RNA m6A cancer. Here, constructed a cell culture model under high concentration glucose tumor evaluate vitro vivo progression. After confirming suppressive HKDC1 antibody tumors, epigenetic alterations (DNA 5mC m6A) baicalin-regulated gene were detected using MS q-PCR. Next, METTL3 gene-regulated (2854 site) was investigated SELECT PCR. Finally, impact other three analogs (baicalein, wogonoside, wogonin) inhibition tested while verifying related mechanism. The results showed suppressed progression vivo. Furthermore, significantly inhibited modification HepG2 mainly targeting site (2854). The m6A-related gene, METTL3, regulated (2854) HKDC1, which also restricted by baicalin. Moreover, study verified METTL3/HKDC1/JAK2/STAT1/caspase-3 pathway cells when exposed concentration. In addition, proven regulate suppress tumors. The findings this revealed regulating METTL3/m6A/HKDC1 axis, might support potential application preventing treating
