Background Platelets are key players in the pathophysiology of coronary artery disease (CAD) and platelet hyperreactivity leads to increased risk developing adverse cardiovascular events. Further, significant changes lipidome occur patients with acute syndrome (ACS) critically regulated lipids lead hyperresponsiveness. Statin treatment is crucial prevention CAD by remodeling lipid metabolism. Objective In this study, we investigate untargeted lipidomics, highlighting between statin-treated naïve patients. Methods We characterized a cohort (n = 105) an lipidomics approach using liquid chromatography coupled mass spectrometry. Results Among annotated lipids, 41 were significantly upregulated patients, whereas 6 downregulated compared The most prominent belong class triglycerides, cholesteryl esters, palmitic acid, oxidized phospholipids, mainly glycerophospholipids untreated A more pronounced effect statin on was observed ACS further highlight dose-dependent influence lipidome. Conclusion Our results reveal that altered embody characteristic mostly compromise glycerophospholipids, which may play role CAD. study contribute understanding softening phenotype.

During the last two decades, constitutive androstane receptor (CAR; NR1I3) has emerged as a master activator of drug- and xenobiotic-metabolizing enzymes transporters that govern clearance both exogenous endogenous small molecules. Recent studies indicate CAR participates, together with other nuclear receptors (NRs) transcription factors, in regulation hepatic glucose lipid metabolism, hepatocyte communication, proliferation toxicity, liver tumor development rodents. Endocrine-disrupting chemicals (EDCs) constitute wide range persistent organic compounds have been associated aberrations hormone-dependent physiological processes. Their adverse health effects include metabolic alterations such diabetes, obesity, fatty disease animal models humans exposed to EDCs. As numerous xenobiotics can activate CAR, its role EDC-elicited gained much interest. Here, we review key features mechanisms xenobiotic-sensing receptor, species differences selectivity ligands, contribution evidence for CAR-dependent EDC action therein.

A fundamental role of pancreatic β-cells to maintain proper blood glucose level is controlled by the Ras superfamily small GTPases that undergo post-translational modifications, including prenylation. This covalent attachment with either a farnesyl or geranylgeranyl group controls their localization, activity, and protein–protein interactions. Small are critical in maintaining homeostasis acting pancreas metabolically active tissues such as skeletal muscles, liver, adipocytes. Hyperglycemia-induced upregulation suggests inhibition these pathways deserves be considered potential therapeutic approach treating T2D. Perspective presents how various points mevalonate pathway might affect protein prenylation functioning diabetes-affected contribute chronic inflammation involved diabetes mellitus (T2D) development. We also demonstrate currently available molecular tools decipher mechanisms linking pathway's enzymes diabetes.

Glucose and cholesterol engage in almost all human physiological activities. As the primary energy substance, glucose can be assimilated converted into diverse essential substances, including cholesterol. Cholesterol is mainly derived from de novo biosynthesis intestinal absorption of diets. It evidenced that glucose/insulin promotes uptake, which have been targeted by several drugs for lipid-lowering, e.g., bempedoic acid, statins, ezetimibe, proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors. Inversely, these lipid-lowering may also interfere with metabolism. This review would briefly summarize mechanisms glucose/insulin-stimulated discuss effect genetic mutations on homeostasis, aiming to help better understand intricate relationship between

Diabetes has emerged as one the leading detrimental factors for human life expectancy worldwide. The disease is mainly considered outcome of dysregulation in glucose metabolism, resulting consistent high concentration blood. At initial stages, diabetes particularly type 2 diabetes, manageable by lifestyle interventions such regular physical activity and diet with less carbohydrates. However, advance stage, intake external insulin dose medicines like metformin are recommended. long-term consumption associated several side effects nausea, vomiting, diarrhoea, lectic acidosis etc., In this scenario, plant-based have shown promising potential prevention treatment diabetes. Berberine bioactive compound present different plant parts berberis family. Biochemical studies that berberine improve sensitivity secretion. Additionally, induces metabolism activating AMPK signaling inhibition inflammation. A series demonstrated antidiabetic at vitro, pre-clinical clinical trials. Many investigations have discovered a connection between statins and magnesium supplements. On one hand, increasing research suggests that chronic hypomagnesemia may be an important factor in the etiology of some metabolic illnesses, including obesity overweight, insulin resistance type 2 diabetes mellitus, hypertension, alterations lipid metabolism, low-grade inflammation. Chronic problems seem to prevented by high Mg intake combined with diet and/or other it is known lower frequency cardiac events, stroke, mortality, not lowering LDL-C, but capacity reduce mevalonate formation. Abstract Background Numerous intrauterine factors may affect the offspring’s growth during childhood. We aimed to explore if maternal and paternal prenatal lipid, apolipoprotein (apo)B apoA1 levels are associated with offspring weight, length, body mass index from 6 weeks eight years of age. This has previously been studied a limited extent. Methods parental negative control study is based on Norwegian Mother, Father Child Cohort Study uses data Medical Birth Registry Norway. included 713 mothers fathers or without self-reported hypercholesterolemia their offspring. Seven metabolites were measured by nuclear magnetic resonance spectroscopy, weight length at 12 time points. Data analyzed linear spline mixed models, results presented as interaction between metabolite (age). Results Higher total cholesterol (TC) level was larger increase in up 8 age (0.03 ≤ P 0.04). Paternal TC not change (0.17 0.25). high-density lipoprotein (HDL-C) lower (0.001 0.005). HDL-C 5 but (0.01 0.03). Parental height (0.07 0.99). Conclusions Maternal compared TC, HDL-C, more strongly consistently childhood, supporting direct effect.
