Numerous studies have indicated broad spectrum pharmacological properties correlations between increased supply the human diet lower prevalence various disorders. The positive effects polyphenols application mostly discussed terms cardiovascular system well-being. However, recent years, they also increasingly mentioned as prophylactic therapeutic factors context neurological diseases, being able to suppress progression such disorders soothe accompanying symptoms. Among over 8000 compounds, that been identified, most widely examined comprise resveratrol, curcumin, luteolin quercetin. This review focuses on vitro assessments, animal models clinical trials, reflecting actual state knowledge, polyphenols' medicinal capabilities epilepsy, demyelinating neurodegenerative diseases central nervous system.

Importance Research is beginning to elucidate the sophisticated mechanisms underlying microbiota-gut-brain-immune interface, moving from primarily animal models human studies. Findings support dynamic relationships between gut microbiota as an ecosystem (microbiome) within (host) and its intersection with host immune nervous systems. Adding this effects on epigenetic regulation of gene expression further complicates strengthens response. At heart inflammation, which manifests in a variety pathologies including neurodegenerative diseases such Alzheimer’s disease, Parkinson’s Multiple Sclerosis (MS). Observations Generally, research date limited has focused bacteria, likely due simplicity cost-effectiveness 16s rRNA sequencing, despite lower resolution inability determine functional ability/alterations. However, omits all other fungi, viruses, phages, are emerging key members microbiome. Much been done pre-clinical and/or small studies more developed parts world. The observed promising but cannot be considered reliable or generalizable at time. Specifically, causal determined currently. More followed by then little MS. data for MS encouraging this. Conclusions relevance While still nascent, interface may missing link, hampered our progress understanding, let alone preventing, managing, putting into remission diseases. Relationships must first established humans, have shown poorly translate complex physiology environments, especially when investigating microbiome where often overly simplistic. The topical administration of in situ hydrogels for ocular pathologies is a promising application strategy providing high effectiveness and patient compliance. Curcumin, natural polyphenol, possesses all the prerequisites successful therapy ophthalmic diseases, but unfortunately its physicochemical properties hurdle practical use. Applying composite thermoresponsive hydrogel formulation embedded with polymer nanoparticles potent to overcome identified drawbacks. In present work we prepared uniform spherical (296.4 ± 3.1 nm) efficiently loaded curcumin (EE% 82.5 2.3%) based on biocompatible biodegradable poly-(lactic-co-glycolic acid). They were thoroughly physicochemically characterized terms FTIR, SEM, TGA, DLS, vitro release following Fickian diffusion (45.62 2.37%), stability over 6 months. Their lack cytotoxicity was demonstrated HaCaT cell lines, potential antioxidant protection also outlined, starting from concentrations as low 0.1 µM reaching 41% at 5 µM. An (17% w/v poloxamer 407 0.1% Carbopol) suitable optimized respect gelation temperature (31.40 0.36 °C), gelling time (8.99 0.28 s) upon tears dilution, gel erosion (90.75 4.06%). Upon curcumin-loaded nanoparticle embedding, appropriate pseudoplastic behavior viscosity 35 °C (2129 24 Pa∙s), 6-fold increase permeation, prolonged h.

Gut microbiota (GM) play a role in the metabolic health, gut eubiosis, nutrition, and physiology of humans. They are also involved regulation inflammation, oxidative stress, immune responses, central peripheral neurotransmission. Aging unhealthy dietary patterns, along with inflammatory responses due to dysbiosis, can lead pathogenesis neurodegenerative diseases, especially Alzheimer’s disease (AD). Although exact mechanism between AD GM dysbiosis is still unknown, recent studies claim that secretions from enhance hallmarks by disturbing intestinal permeability blood–brain barrier via microbiota–gut–brain axis. Dietary polyphenols secondary metabolites plants possess anti-oxidative anti-inflammatory properties ameliorate enhancing abundance beneficial bacteria. Thus, modulation prevent treat other diseases. This review summarizes AD. Further, it provides an overview on ability modulate inflammation against

Alzheimer's disease (AD) is characterized by the progressive degeneration of neuronal cells. With increase in aged population, there a prevalence irreversible neurodegenerative changes, causing significant mental, social, and economic burden globally. The factors contributing to AD are multidimensional, highly complex, not completely understood. However, it widely known that aging, neuroinflammation, excessive production reactive oxygen species (ROS), along with other free radicals, substantially contribute oxidative stress cell death, which inextricably linked. While undeniably important AD, limiting radicals ROS levels an intriguing potential strategy for deferring process neurodegeneration alleviating associated symptoms. Therapeutic compounds from natural sources have recently become increasingly accepted been effectively studied treatment. These phytocompounds available multitude holistic therapeutic efficiencies treating owing their antioxidant, anti-inflammatory, biological activities. Some these also function stimulating cholinergic neurotransmission, facilitating suppression beta-site amyloid precursor protein-cleaving enzyme 1, α-synuclein, monoamine oxidase proteins, deterring occurrence AD. Additionally, various phenolic, flavonoid, terpenoid extensively described as palliative agents progression. Preclinical studies shown involvement modulating cellular redox balance minimizing formation, displaying them antioxidant neuroprotective abilities.
