Functions of selenium are diverse as antioxidant, anti-inflammation, increased immunity, reduced cancer incidence, blocking tumor invasion and metastasis, further clinical application treatment with radiation chemotherapy. These functions mostly related to oxidation reduction mechanisms metabolites. Hydrogen selenide from selenite, methylselenol (MSeH) Se-methylselenocyteine (MSeC) methylseleninicacid (MSeA) the most reactive metabolites produced oxygen species (ROS); furthermore, these may involve in oxidizing sulfhydryl groups, including glutathione. Selenite also reacted glutathione produces hydrogen via selenodiglutathione (SeDG), which induces cytotoxicity cell apoptosis, ROS production, DNA damage, adenosine-methionine methylation cellular nucleus. However, a more pronounced effect was shown subsequent sodium selenite chemotherapy therapy. High doses were effective increase therapy chemotherapy, reduce side effects drug resistance. Leishmaniasis affects more than 12 million humans globally and a further 1 billion people are at risk in leishmaniasis endemic areas. This study was undertaken to evaluate potential some organoselenium compounds by quantitative structure-activity relationship (QSAR) modeling, molecular docking, analysis, dynamic (MD) simulation. built QSAR model validated (R2train = 0.8646, R2test 0.8864, Q2 0.5773) predicted inhibitory activity (pIC50) values newly designed were higher that template (Compound 6). analogues (6a, 6b, 6c) showed good binding interactions target protein (Pyridoxal kinase, PdxK) while also presenting excellent drug-likeness profiles. results density functional theory, MD simulation, mechanics generalized Born surface area (MM/GBSA) analyses suggest favourability stability PdxK, comparing favourably well reference (Pentamidine).
