Autism spectrum disorder (ASD) imposes a significant burden on both patients and society. Amino acid metabolism abnormalities are particularly relevant to ASD pathology due their crucial role in neurotransmitter synthesis, synaptic function, overall neurodevelopment. This study aims explore the association between amino metabolic severity of by analyzing concentrations blood children with ASD. Fasting peripheral samples were collected from 344 ASD, measured using liquid chromatography-tandem mass spectrometry (LC-MS/MS) while strictly following quality control measures. The was evaluated logistic regression restricted cubic spline (RCS) analysis. ROC (receiver operating characteristic) curve, decision curve analysis (DCA), calibration used construct validate predictive models nomograms, thereby assessing performance. Multivariate showed that aspartic (OR = 1.037, 95% CI: 1.009-1.068, P 0.01), glutamic 1.009, 1.001-1.017, 0.03), phenylalanine 1.036, 1.003-1.072, 0.04), leucine/isoleucine 1.021, 1.006-1.039, 0.01) significantly positively correlated On other hand, tryptophan 0.935, 0.903-0.965, < valine 0.987, 0.977-0.997, negatively RCS further revealed nonlinear relationship acid, proline, risk combined model achieved an AUC (area under curve) 0.806, indicating high diagnostic accuracy. Autism spectrum disorder (ASD) is a neurodevelopmental pathology characterized by the impairment of social interaction, difficulties in communication, and repetitive behaviors. Alterations metabolism amino acids have been reported. We performed chromatographic analysis fecal acids, ammonium, biogenic amines, gamma aminobutyric acid (GABA) Tunisian autistic children from 4 to 10 years, results were compared with their siblings (SIB) general population (GP). ASD presented significantly higher levels than SIB GP; differences being more pronounced younger (4-7 years) older (8-10 individuals whereas no changes found for remaining compounds. Lower histidine only difference related severe symptoms autism (CARS scale). A linear discriminant (LDA) based on profiles clearly separated ASD, SIB, GP at 7 years but not advanced age years), evidencing alterations children. The relationship needs deeper research integrating blood analytical parameters, brain metabolism, intestinal microbiota. In last few years, it has been hypothesized that gut microbiota dysbiosis is strongly implicated in aetiopathogenesis of ASD. Recently, new studies have suggested may be involved neurological and behavioural defects caused by pesticides, ASD symptoms. This review highlights available from recent animal human on relationship between potential disturb intestinal homeostasis, The mechanisms through which trigger ASD-like behaviours induced via altered production bacterial metabolites (short chain fatty acids, lipids, retinol, amino acid) are also described.
