Ferroptosis, an emerging paradigm of programmed cellular necrosis posited in recent years, manifests across a spectrum maladies with profound implications for human well-being. Numerous investigations substantiate that modulating ferroptosis, whether through inhibition or augmentation, plays pivotal role the etiology and control numerous age-related afflictions, encompassing neurological, circulatory, respiratory, other disorders. This paper not only summarizes regulatory mechanisms but also discusses impact ferroptosis on biological processes aging its diseases. Furthermore, it scrutinizes therapeutic strides addressing aging-related conditions modulation ferroptosis. The consolidates existing knowledge potential applications ferroptosis-related pharmacotherapies envisages translational prospects ferroptosis-targeted interventions clinical paradigms.

Abstract Cortical bone loss is intricately associated with ageing and coincides iron accumulation. The precise role of ferroptosis, characterized by overload lipid peroxidation, in senescent osteocytes remains elusive. We found that ferroptosis was a crucial mode osteocyte death cortical during ageing. Using single‐cell transcriptome analysis, we identified activating transcription factor 3 (ATF3) as critical driver ferroptosis. Elevated ATF3 expression promotes uptake upregulating transferrin receptor 1 while simultaneously inhibiting solute carrier family 7‐member 11‐mediated cystine import. This process leads to an culminating Importantly, inhibition aged mice effectively alleviated the mitigated mass loss. Taken together, our findings establish pivotal older adults, providing promising prevention treatment strategies for osteoporosis fractures.

Neurodegenerative disorders, particularly Alzheimer’s disease (AD), remain a great challenge regarding the finding of effective treatment, one main reason being incomplete understanding their etiology. With many intensely debated hypotheses, newer approach based on impact iron imbalance in sustaining neurodegeneration central nervous system becomes increasingly popular. Altered homeostasis leads to increased accumulation specific brain areas, explaining clinical picture AD patients. Moreover, growing evidence sustains significant metabolism relationship other pathological processes encountered AD-affected brain, such as amyloidogenic pathway, chronic inflammation, or oxidative stress. In this context, mini-review aims summarize novel data from continuously expanding literature topic didactic manner. Thus, first part, authors briefly highlight most relevant aspects related absorption, transport, regulation, and elimination at cerebral level, focusing role blood-brain barrier concept ferroptosis. Subsequently, currently available chelation therapies are discussed, including an overview trials topic. final latest results vitro vivo studies, new research directions suggested enhance development antidementia therapies.

Abstract Heavy metals influence the development of several health conditions, including inflammation and cancer. This study investigates relationship between heavy metal concentrations in plasma urine, presence benign prostatic hyperplasia (BPH), precancerous lesions (PL), prostate cancer (PC). The age, total PSA levels, hemoglobin concentrations, monocyte-to-lymphocyte ratio (MLR) is also analyzed across three groups: BPH, PL, PC. Our findings reveal significant differences vanadium antimony plasma, suggesting a potential role disease pathophysiology. Notably, lower are associated with an increased risk PC, while significantly higher PL group. Regression analysis further supports association highlighting copper as biomarkers or therapeutic targets for health. explores impact lead exposure on risk, revealing urine concentration These underscore complex interaction emphasizing need research to elucidate underlying mechanisms explore interventions.
