Allergic contact dermatitis (ACD) is a skin condition characterized by inflammation resulting from hypersensitivity upon with certain allergens. Although ACD an immune-mediated pathomechanism, the involvement of nervous system in this has increasingly been considered, particularly amplification and persistence inflammation. This paper aims to present comprehensive overview mechanisms involved neurogenic ACD, focusing on role sensory neurons, release neuropeptides, their interaction immune cells, potential therapeutic implications related pathways, diversified age gender. Innovative therapies for including topical formulations, may target mass-bound X2 G-protein-coupled receptor (MRGPRX2) endocannabinoid systems.

Abstract Botulinum toxin type A (BoNT) has emerged as a potential alternative to conventional therapies many debilitating chronic diseases characterised by inflammatory states. However, the biological rationale remains ambiguous. Our review aimed systematically assessed which biochemical biomarkers have been reported in clinical research evaluate BoNT analgesic and mood-lifting effects head neck conditions related inflammation. We searched databases registries between inception September 29, 2023. Of nine included studies, there were concerns about risk of bias for six studies. The leading biomarker with five studies was calcitonin gene-related peptide (CGRP), followed serotonin two Oxidative stress only one study. Several important players processes different immune cell classes evaluated four There trial measuring changes beta Tubulin SNAP-25, another study evaluating cutaneous neuropeptide substance-P. After BoNT, significant effect including decrease plasma levels CGRP migraine trigeminal neuralgia; when collected from human tears refractory intractable dry eye disease increase peripheral blood platelets painful cervical dystonia associated depression anxiety; concentration markers oxidative damage proteins antioxidant power; expression gene sets involved pathways cells periosteum metalloproteinase-9 molecule tears. seems affect some present conditions. certainty evidence found very low moderate. This is registered on PROSPERO (CRD42023432131).

Rosacea is a common chronic inflammatory condition primarily affecting middle-aged women. It presents with flushing, erythema, telangiectasia, papules, pustules, phymatous changes, and ocular involvement. Although typically grouped into four subtypes-erythematotelangiectatic, papulopustular, ocular, phymatous-overlapping features often favor phenotypic diagnostic approach. Neurogenic rosacea (NR) has emerged as distinct subgroup featuring distinguishing such peripheral facial severe burning stinging sensations, resistance to standard therapies. Recent insights the pathophysiology of NR propose neural dysregulation main driver condition. Specifically, activation TRP channels at cutaneous sensory nerve endings in dermis triggers release vasoactive peptides, driving neuroinflammation resulting stinging. Additionally, there marked association neuropsychiatric comorbidities, which would further mediate pathogenesis In line this pathophysiological model, fails respond conventional treatments. Instead, patients benefit more from antidepressants neuroleptic agents that help modulate neuronal activity alleviate symptoms. This review explores summarizes scientific evidence regarding new on disease pathogenesis, clinical manifestations, proposed treatments for NR.
