Successful engineering of functional salivary glands necessitates the creation cell‐instructive environments for ex vivo expansion and lineage specification primary human gland stem cells (hS/PCs). Herein, basement membrane mimetic hydrogels are prepared using hyaluronic acid, cell adhesive peptides, hyperbranched polyglycerol (HPG), with or without sulfate groups, to produce “hyperGel+” “hyperGel”, respectively. Differential scanning fluorescence experiments confirm ability sulfated HPG precursor stabilize fibroblast growth factor 10. The nanoporous, cytocompatible, cell‐permissive, enabling development multicellular hS/PC spheroids in 14 days. incorporation species hydrogel enhances proliferation. Culture hS/PCs hyperGel+ presence a Rho kinase inhibitor Y‐27632 (Y‐27) leads central lumen, increases expression acinar marker aquaporin‐3 at transcript level ( AQP3 ), decreases ductal keratin 7 both KRT7 ) protein levels (K7). Reduced transforming beta (TGF‐β) targets SMAD2/3 is also observed Y27‐treated cultures, suggesting attenuation TGF‐β signaling. Thus, cooperates Rho‐associated promote lumened enhanced markers.

Upadacitinib, a selective Janus associated kinase 1 (JAK-1) inhibitor, can be prescribed particularly for the clinical treatment with Crohn's disease or rheumatoid arthritis. It is clinically observed that upadacitinib has been found potential therapeutic effectiveness on Sjogren's syndrome (SS). However, anti-SS targets and mechanisms involved in remain uninvestigated.

Abstract Background Sjogren's syndrome (SS) is a complex autoimmune disease influenced by genetics, yet its genetic underpinnings remain elusive. This study investigates the correlation and potential causative link between cytokine cycling levels SS. Methods Genome-wide association studies (GWAS) were conducted with 8,293 14,824 European participants to identify cytokines. The GWAS dataset for SS, comprising 368,028 individuals of ancestry (2,495 cases 365,533 controls), was sourced from Finnish biological sample library. Single nucleotide polymorphisms (SNPs) associated SS identified using Linkage disequilibrium score (LDSC) regression Mendelian randomization (MR) analysis. inverse variance weighted (IVW) method primary analytical approach. Additional methods including MR Egger, Weighted median, mode employed robustness assessment. Heterogeneity testing, horizontal pleiotropy Steiger testing sensitivity Reverse analysis performed assess reverse causal relationship Results LDSC 46 cytokines bidirectional IVW revealed significant associations genetically predicted IL10RB (P = 0.019, OR 1.138, 95% CI: 1.021–1.267) CXCL11 0.015, 1.269, 1.048–1.537) increased risk. absence heterogeneity in underscores these findings. Conclusion suggests particularly through cycles. Further research warranted elucidate mechanisms which influence

Introduction. A rare extraglandular manifestation of Sjögren's syndrome is presented, mimicking idiopathic intracranial hypertension. Case Report. young eutrophic female, complained persistent headache, followed by diplopia and bilateral sixth nerve paresis. Neuroimaging investigation showed classic signs As the patient had a normal weight, prompted evaluation secondary causes, which revealed positive anti-SSA/Ro anti-La/SSB antibodies. salivary gland biopsy later confirmed diagnosis syndrome. Xerostomia dysphagia developed weeks later. Conclusion. This case report important to raise awareness about an atypical neurological
