We genetically manipulated HaCaT cells, a spontaneously immortalised normal keratinocyte cell line, to stably express two different coloured luciferase reporter genes, driven by interleukin 8 ( IL‐8 ) and ubiquitin‐C UBC promoters, respectively. Subsequently, we generated three‐dimensional (3D) skin‐like in vitro composite (SLIC) utilising these with the objective of monitoring bioluminescence emitted from SLIC. This SLIC was on non‐woven silica fibre membranes differentiation medium. Excess exogenous supplementation of D-galactose (D-gal), a monosaccharide and reducing sugar, generates reactive oxygen species (ROS), leading to cell damage death. ROS accumulation is critical in aging. Therefore, D-gal-induced aging mouse models are used studies. Herein, we evaluated D-gal's effect on neonatal testis development using an vitro organ culture method. Mouse testicular fragments (MTFs) derived from testes (postnatal day 5) were cultured with 500 mM D-gal for 5 days. D-gal-treated MTFs showed significantly increased decreased expression undifferentiated differentiated germ markers, respectively, substantial reduction meiotic cells. In D-gal-exposed MTFs, levels Sertoli markers (Sox9 Wt1) increased, while those StAR 17β-HSD3, whose expressions abundant D-Gal treated adult Leydig cells, decreased. Additionally, the enzyme 3 β-HSD1, essential steroidogenesis was reduced compared that controls.D-gal Bad, Bax, cleaved caspase-3 -8. Via oxidative stress MTF. An efficient method to synthesize functionalized allyl isothiocyanates in moderate yields has been demonstrated, based on the isomerization of Morita-Baylis-Hillman adduct-derived allylic thiocyanates. This methodology offers obvious advantages such as easy purification, atom economy, and a broad scope substrates.
