New drugs take effect at level central nervous system, gastrointestinal tract, adipose tissue, kidneys, liver, pancreas, skeletal muscles. The safety gene therapy being evaluated, potential special methods drug delivery target tissues studied. Randomized studies show that effectiveness a number new pharmacological weight carbohydrate metabolism normalization already almost comparable bariatric surgery. Abstract ObjectiveOxidative stress decreases the ability of β-cells to secrete insulin through glucolipotoxicity pancreatic islets. Flavonoids modulate and amylin secretion mainly antioxidant activities. Coumarins isomers flavonoids have direct effects on cardiovascular system, not linked This study aimed investigate 5,7-dimethoxycoumarin (Citropten) fractions present in grapefruit peel secretions normal male Wistar rats.MethodsMethanol extract peels was fractionated using vacuum assisted liquid chromatography with n-Hexane, Ethyl acetate Methanol. Gas Chromatography Mass Spectrometry analysis reported ethyl fraction highest concentration (85.66%) 5,7-dimethoxycoumarin. Intraperitoneal Glucose Tolerance Test performed 5 sets rats receiving intraperitoneally: 1) negative control, 1ml sterile water 2) positive 0.2mg/kg glimepiride, 3) containing 10mg/kg, 20mg/kg 5,7-dimethoxycoumarin, 4) methanol 5a)1ml H2O2(0.6%, 6%) plus 5b)1000mg/kg Vitamin C 5,7-dimethoxycoumarin.ResultsResults showed 10mg/kg had comparable plasma glucose control indirect effect unlike acting glimepiride; less effective. Both 5,7- dimethoxycoumarin induced extrapancreatic synthesis secretion.Conclusion5,7-dimethoxycoumarin will find special applications management obesity, diabetics chronic complications. Since overall regulation is achieved synergy, attention should be shifted from insulin-based amylin-based therapy. There need focus natural compounds that stimulate release.
