Abstract Osteoarthritis (OA) is a common degenerative joint disease with complex risk factors and an unclear pathogenesis. The onset insidious, mild early symptoms, the progression irreversible. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovial inflammation, joint destruction, and systemic complications. The nucleotide-binding domain, leucine-rich repeat family, pyrin domain-containing-3 (NLRP3) inflammasome plays pivotal role in RA pathogenesis driving the release of pro-inflammatory cytokines exacerbating oxidative stress. Recent studies identified methyl canthin-6-one-2-carboxylate (Cant) as potential therapeutic agent that modulates NLRP3 pathway. This review explores mechanistic Cant treatment, particularly its effect on stress, macrophages, inflammatory signaling pathways. Additionally, we discuss alternative complementary approaches, such gut microbiota modulation mesenchymal stem cell-based therapies, management RA. Although preliminary findings suggest exhibits promising anti-inflammatory effects, further preclinical clinical are necessary to validate efficacy. Future research should focus optimizing dosage, exploring combination elucidating broader implications targeting for treatment.

ABSTRACT Disrupted intestinal epithelial barrier function has been proposed to be integral rheumatoid arthritis (RA) progression and pathogenesis. To further define the molecular pathways in synovial inflammation response of tissues, we have used a rat model mono-joint inflammatory arthritis, induced by intra-articular injection Complete Freund’s adjuvant (CFA). Abstract Background The knee joint is one of the most frequently affected joints in osteoarthritis (OA). However, specific connection between gastric diseases and occurrence (KOA) currently unclear. objective this study to explore potential association KOA using China Health Retirement Longitudinal Study (CHARLS) database. Methods A total 1053 subjects with complete information 2018 from CHARLS database were included. First, baseline characteristics these (covariates such as gender, residence, smoke) compared.
