We aimed to systematically evaluate and analyze the clinical efficacy of oral probiotics in treatment knee osteoarthritis (KOA) based on theory "gut-joint axis." searched PubMed, The Cochrane Library, Embase, China Knowledge Network (CNKI), Wanfang Database, Wipro Database (CQVIP) databases for randomized controlled trials KOA. literature was organized by Note express software, quality included evaluated according systematic evaluation method, meta-analysis performed using RevMan 5.4 software. Five with 694 participants were this study, results showed that observation group experienced significant reductions Western Ontario McMaster Universities Osteoarthritis Index total score, visual analog high-sensitivity C-reactive protein level compared control group, but did not show differences improvement stiffness regulation body weight. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovial inflammation, joint destruction, and systemic complications. The nucleotide-binding domain, leucine-rich repeat family, pyrin domain-containing-3 (NLRP3) inflammasome plays pivotal role in RA pathogenesis driving the release of pro-inflammatory cytokines exacerbating oxidative stress. Recent studies identified methyl canthin-6-one-2-carboxylate (Cant) as potential therapeutic agent that modulates NLRP3 pathway. This review explores mechanistic Cant treatment, particularly its effect on stress, macrophages, inflammatory signaling pathways. Additionally, we discuss alternative complementary approaches, such gut microbiota modulation mesenchymal stem cell-based therapies, management RA. Although preliminary findings suggest exhibits promising anti-inflammatory effects, further preclinical clinical are necessary to validate efficacy. Future research should focus optimizing dosage, exploring combination elucidating broader implications targeting for treatment.

ABSTRACT Disrupted intestinal epithelial barrier function has been proposed to be integral rheumatoid arthritis (RA) progression and pathogenesis. To further define the molecular pathways in synovial inflammation response of tissues, we have used a rat model mono-joint inflammatory arthritis, induced by intra-articular injection Complete Freund’s adjuvant (CFA). The predominant single into knee joint resulted rapid reproducible formation fibrotic myeloid-infiltrated pannus.
