Every 25th death worldwide is associated with liver pathology. The development of novel approaches to diseases therapy and protocols for maintaining the vital functions patients on transplant waiting list are urgently needed. Resident mesenchymal stem cells (MSCs) play a significant role in supporting tissue integrity improve condition after infusion. However, it remains unclear whether MSCs isolated from chronically inflamed livers similar their basic cellular properties obtained healthy livers. We applied large array tests compare resident apparently normal fibrosis, cirrhosis, viral hepatitis. Chronic inflammatory environment did not alter major characteristics MSCs, including expression MSC markers, cell adhesion molecules, hallmarks senescence, as well proliferation, migration, secretome. Only some immune checkpoints toll-like receptors was different. Evidently, unchanged present human even at late stages diseases. These can be used starting material therapies

Metabolic-Associated Fatty Liver Disease (MAFLD) is a clinical-pathological scenario that occurs due to the accumulation of triglycerides in hepatocytes which considered significant cause liver conditions and contributes an increased risk death worldwide. Even though possible causes MAFLD can involve interaction genetics, hormones, nutrition, lifestyle (diet sedentary lifestyle) most influential factor developing this condition. Polyphenols comprise many natural chemical compounds be helpful managing metabolic diseases. Therefore, aim review was investigate impact oxidative stress, inflammation, mitochondrial dysfunction, role polyphenols MAFLD. Some reverse part damage related or among them are anthocyanin, baicalin, catechin, curcumin, chlorogenic acid, didymin, epigallocatechin-3-gallate, luteolin, mangiferin, puerarin, punicalagin, resveratrol, silymarin. These have actions reducing plasma enzymes, body mass index, waist circumference, adipose visceral indices, lipids, glycated hemoglobin, insulin resistance, HOMA index. They also reduce nuclear factor-KB (NF-KB), interleukin (IL)-1β, IL-6, tumor necrosis factor-α (TNF-α), blood pressure, fat content, steatosis fibrosis. On other hand, they improve HDL-c, adiponectin levels, fibrogenesis markers. results show promising prevention treatment

Vitamin A is a crucial nutrient renowned for its role in visual health and cellular regulation. Its derivatives influence cell differentiation, proliferation, tissue homeostasis, making them significant cancer research due to their effects on both normal tumour cells. This review explores the intricate relationship between vitamin metabolism extracellular matrix (ECM) cancer. The ECM profoundly affects behaviour, including invasion, metastasis. Alterations can facilitate progression, have shown potential modulating these changes. Through transcriptional regulation, impacts components metalloproteinases, influencing dynamics. highlights of as adjunctive agents therapy. Despite promising laboratory findings, clinical application remains limited challenges translating into therapeutic outcomes. Future should focus modulation retinol within tumours development targeted therapies enhance treatment efficacy improve patient prognosis.

Cirrhotic portal hypertension, the most prevalent and clinically significant complication of liver cirrhosis, manifests as elevated venous pressure is associated with severe complications. Although much research on mechanisms hypertension has focused fibrosis, less attention been given to role intrahepatic extrahepatic vascular dysfunction, particularly respect vasculature. While hepatic fibrosis in cirrhotic undeniable, underlying involving vasculature are highly complex. Sinusoidal capillarization endothelial dysfunction contribute increased resistance. Hemodynamic changes circulation, including splanchnic vasodilation hyperdynamic play a development hypertension. Additionally, therapeutic strategies targeting these diverse, improvement sinusoidal microcirculation, therapies stellate cells activation, pharmacological modulation systemic tone. Therefore, this review, we will discuss vascular-related treatment progress cirrhosis provide new theoretical basis practical guidance for clinical treatment.

Stress-induced liver injury, resulting from acute or chronic stress, is associated with oxidative stress and inflammation. The endocannabinoid system, particularly cannabinoid receptor 2 (CB2R), plays a crucial role in damage. However, there are currently no clinical drugs targeting CB2R for diseases. Cannabidiol (CBD), agonist, possesses anti-inflammatory antioxidant properties. This study aims to investigate the pharmacological effects of CBD mouse model stress-induced injury. We employed injury evaluate protective CBD. Assessments included histopathological analysis, cytokine detection via ELISA, protein expression analysis using immunohistochemistry Western blot, gene transcription differential analysis. Transmission electron microscopy was utilized observe mitochondrial morphology. Additionally, we examined levels CB2R, SLC7A11, α-SMA, ACSL4 proteins elucidate mechanisms underlying CBD's effects. exhibited significant against mice. Decreases function indicators (including Aspartate Aminotransferase (AST) Alanine (ALT)) inflammatory cytokines (such as IL-1β Tumor Necrosis Factor-alpha (TNF-α)) were observed. enhanced reduced α-SMA levels, mitigating fibrosis. It also decreased increased SOD GSH-Px activities, upregulated SLC7A11 expression. Furthermore, improved morphology, indicating reduction cell death. activates CB2R/α-SMA pathway modulate inflammation Through SLC7A11/ACSL4 signaling pathway, alleviates enhances reduces These findings provide theoretical basis potential application prevention treatment

Background and Objectives: Metabolic syndrome (MetS) is a condition marked by complex array of physiological, biochemical, metabolic abnormalities, including central obesity, insulin resistance, high blood pressure, dyslipidemia (characterized elevated triglycerides reduced levels high-density lipoproteins). The pathogenesis develops from the accumulation lipid droplets in hepatocyte (steatosis). This accumulation, genetically predisposed subjects with other external stimuli (intestinal dysbiosis, caloric diet, physical inactivity, stress), activates production pro-inflammatory molecules, alter autophagy, turn on activity hepatic stellate cells (HSCs), provoking low grade chronic inflammation fibrosis. associated significantly increased risk developing type 2 diabetes mellitus (T2D), cardiovascular diseases (CVD), vascular, renal, pneumologic, rheumatological, sexual, cutaneous syndromes overall mortality, rising five- to seven-fold for T2DM, three-fold CVD, one half–fold all-cause mortality. purpose this narrative review examine as “systemic disease” its interaction major internal medicine conditions such diabetes, renal failure, respiratory failure. It essential practitioners approach widespread “holistic” rather than fragmented manner, particularly Western countries. Additionally, it important be aware non-invasive tools available assessing condition. Materials Methods: We conducted an exhaustive search PubMed up July 2024, focusing terms related pathologies (heart, Lung (COPD, asthma, pulmonary hypertension, OSAS) kidney rheumatological (osteoarthritis, rheumatoid arthritis), endocrinological, sexual neoplastic risks. was managed accordance PRISMA statement. Finally, we selected 300 studies (233 papers first strategy 67 second one). Our included that provided insights into techniques evaluating liver fibrosis steatosis. Studies were not humans, published languages English, or did assess changes heart failure excluded. Results: findings revealed clear correlation between all above described, indicating assessments steatosis could potentially serve markers severity progression diseases. Conclusions: multisystem disorder impacts organs beyond disrupts functioning various organs. Notably, linked higher incidence diseases, independent traditional factors. Non-invasive allow clinicians evaluate risk. ability may open new diagnostic, therapeutic, prognostic avenues managing complications, disease, which leading cause death these patients.
