From Polydeoxyribonucleotides (PDRNs) to Polynucleotides (PNs): Bridging the Gap Between Scientific Definitions, Molecular Insights, and Clinical Applications of Multifunctional Biomolecules

Polydeoxyribonucleotides
(PDRNs)
and
polynucleotides
(PNs)
are
similar
DNA-derived
biopolymers
that
have
garnered
significant
scientific
attention
since
the
1990s
for
their
potential
applications
in
wound
healing
skin
rejuvenation.
These
exhibit
a
broad
molecular
weight
(MW)
range,
typically
spanning
from
50
to
1500
kDa.
However,
recent
studies
expanded
this
range
encompass
fragments
as
small
1
kDa
large
10,000
Clinically,
PDRN/PN
formulations,
commercially
available
various
galenic
forms
(gels,
creams,
serums,
masks,
injectables),
demonstrated
promising
effects
significantly
promoting
regeneration,
reducing
inflammation,
improving
texture,
preventing
scar
formation,
mitigating
wrinkles.
Importantly,
despite
widespread
use
cosmetology
aesthetic
dermatology,
interchangeable
of
terms
“PDRN”
“PN”
literature

describe
polymers
varying
lengths)
has
led
considerable
confusion
within
medical
communities.

address
ambiguity,
narrative
review
proposes
standardized
structure-based
nomenclature

Skin
aging
is
a
complex
and
multifactorial
process
influenced
by
both
intrinsic
extrinsic
factors.
The
periorbital
area
of
the
face
particularly
susceptible
to
premature
signs
due
its
delicate
skin
structure,
major
concern
for
many
individuals.
While
hyaluronic
acid
(HA)-based
dermal
filler
products
are
commonly
used
rejuvenation,
novel
approaches
effectively
locally
address
visible
available.
This
study
aimed
investigate
Innovyal
Regenerative
Action
(IRA),
an
injectable
polynucleotide-HA
(PN-HA)
regenerative
designed
periocular
prejuvenation.
Firstly,
PN-HA
was
compared
other
commercially
available
HA-based
dermbooster
(Profhilo®,
Suisselle
Cellbooster®
Glow,
NCTF®
135
HA)
in
terms
rheological
properties,
vitro
antioxidant
capacity,
total
collagen
production
stimulation
human
fibroblasts.
Secondly,
clinical
effects
IRA
were
evaluated
two
case
reports
(monotherapy
prejuvenation).
It
shown
that
outperformed
comparators
relative
behavior
(biophysical
attributes
normalized
polymer
contents),
activity
(CUPRAC,
FRAP,
ORAC
assays),
as
well
level
induction
(72-h
fibroblast
model).
Generally,
results
this
provided
mechanistic
preliminary
insights
into
potential
benefits
cutaneous
treatment.

Recently,
various
biocompatible

microspheres

Polydeoxyribonucleotide
(PDRN),
derived
from
chum
salmon
(Oncorhynchus
keta),
is
a
mixture
of
hydrolyzed
DNA
fragments
used
in
various
clinical
applications.
Its
therapeutic
value
stems
its
ability
to
promote
wound
healing
by
upregulating
growth
factors
like
VEGF,
FGF,
and
HIF-1.
However,
PDRN's
regenerative
properties
raise
concerns
about
potential
misuse
sports.
Studies
suggest
it
may
enhance
athletic
performance
stimulating
muscle
growth,
recovery,
endurance
through
mechanisms
such
as
satellite
cell
activation,
angiogenesis,
anti-inflammatory
effects.
These
performance-enhancing
effects
could
be
considered
gene
or
doping,
prohibited
the
World
Anti-Doping
Agency
(WADA).
To
address
this
concern,
we
developed
sensitive
specific
detection
method
for
PDRN
based
on
CRISPR-Cas12a
system.

targets

without
prior
