Excessive intake of free fatty acids (FFAs), especially saturated acids, can lead to atherosclerosis and increase the incidence cardiovascular diseases. FFAs also contribute obesity, hyperlipidemia, nonalcoholic liver disease. Palmitic acid (PA) is human plasma’s most abundant acid. It often used study toxicity caused by in different organs, including vascular lipotoxicity. Fatty overload induces endothelial dysfunction through various molecular mechanisms. Endothelial alters homeostasis reducing vasodilation increasing proinflammatory prothrombotic states. linked atherosclerosis, which leads coronary artery disease, peripheral stroke. In this review, we summarize latest studies, revealing mechanism acid-induced dysfunction, targeting insulin resistance, reactive oxygen species, inflammation, programmed cell death, ER stress, mitochondrial dysfunction. Meanwhile, review provides new strategies perspectives for preventing impact diseases on health relevant mechanism.
