The Crucial Role of the Blood–Brain Barrier in Neurodegenerative Diseases: Mechanisms of Disruption and Therapeutic Implications

The
blood-brain
barrier
(BBB)
is
a
crucial
structure
that
maintains
brain
homeostasis
by
regulating
the
entry
of
molecules
and
cells
from
bloodstream
into
central
nervous
system
(CNS).
Neurodegenerative
diseases
such
as
Alzheimer's
Parkinson's
disease,
well
ischemic
stroke,
compromise
integrity
BBB.
This
leads
to
increased
permeability
infiltration
harmful
substances,
thereby
accelerating
neurodegeneration.
In
this
review,
we
explore
mechanisms
underlying
BBB
disruption,
including
oxidative
stress,
neuroinflammation,
vascular
dysfunction,
loss
tight
junction
integrity,
in
patients
with
neurodegenerative
diseases.
We
discuss
how
breakdown
contributes
neurotoxicity,
abnormal
accumulation
pathological
proteins,
all
which
exacerbate
neuronal
damage
facilitate
disease
progression.
Furthermore,
potential
therapeutic
strategies
aimed
at
preserving
or
restoring
function,
anti-inflammatory
treatments,
antioxidant
therapies,
approaches
enhance
integrity.

Using

such

we
analyzed
articles
topics
as:
POCD,
GSH,
microglia,
astrocyte,
oligodendrocyte,
ferroptosis,
BDNF,
Neuroinflammation,
stress.
The
above
searched
in
databases
using
Boolean
operations.
We
included
original
articles,
reviews
other
article
types
books.
According
reviewed
literature,
GSH
treatment
for
Specific
targeted
therapies
POCD
still
sparse,
therefore,
implementation
preventive
strategies
appears
remain
optimal
attitude.

Abstract
Neuroinflammation
plays
an
important
role
in
the
pathogenesis
of
various
central
nervous
system
(CNS)
diseases.
The
NLRP3
inflammasome
is
intracellular
multiprotein
complex
composed
innate
immune
receptor
NLRP3,
adaptor
protein
ASC,
and
protease
caspase-1.
activation
can
induce
pyroptosis
release
proinflammatory
cytokines
IL-1β
IL-18,
thus
playing
a
inflammatory
responses.
Recent
studies
have
revealed
that
activated
brain
to
neuroinflammation,
leading
further
neuronal
damage
functional
impairment,
contributes
pathological
process
neurological
diseases,
such
as
multiple
sclerosis,
Parkinson’s
disease,
Alzheimer’s
stroke.
