LEAP2 (liver expression antimicrobial peptide 2), is an widely found in vertebrates and mainly expressed liver. plays a vital role host innate immunity. In teleosts, number of homologs have been reported, but their vivo effects on defense are still limited. this study, homolog (SsLEAP2) was identified from black rockfish, Sebastes schlegelii, its structure, biological functions were analyzed. The results showed that the open reading frame SsLEAP2 300 bp, with 5’- untranslated region (UTR) 375 bp 3’ - UTR 238 bp. deduced amino acid sequence shares highest overall identity (96.97%) umbrosus. possesses conserved features, including signal sequence, prodomain mature peptide, which four well-conserved cysteines formed two intrachain disulphide domain. liver could be induced by experimental infection Listonella anguillarum, Edwardsiealla piscicida Rock bream iridovirus C1 (RBIV-C1). Recombinant (rSsLEAP2) purified Escherichia coli able to bind various Gram-positive Gram-negative bacteria. Further analysis rSsLEAP2 enhance respiratory burst activity, induce immune genes macrophages; as well promote proliferation chemotactic peripheral blood lymphocytes (PBLs). experiments indicated overexpression inhibit bacterial infection, increase level genes; conversely, knock down promoted decreased genes. Taken together, these suggest novel apparent antibacterial activity immunoregulatory property, thus critical against pathogens invasion.

Antimicrobial peptides (AMPs) are promising molecules in diverse fields, including aquaculture. AMPs possess lytic effects on a wide range of pathogens, resulting potential replacement for traditional antimicrobials In addition, they also have modulatory host immune responses. ABSTRACT Dipeptides were constructed using hydrophobic amino acid residues following AMP prediction. After that Boc‐modification was performed on the screened peptides and finally Boc‐Phe‐Trp‐OMe Boc‐Trp‐Trp‐OMe synthesized. Even though no inhibition zones observed in agar well diffusion assays, minimum inhibitory concentration (MIC) analysis revealed anti‐bacterial activity against both Gram‐positive Gram‐negative bacteria, with MIC 90 ranging from 230 to 400 μg/mL. The crystal violet assay confirmed dipeptides' biofilm eradication disruption capabilities. Furthermore, membrane permeabilization assays indicated outer inner permeabilization, while SEM formation of fibril spherical nanostructures, likely contributing this effect. also exhibited resistance protein adsorption, non‐cytotoxicity, non‐hemolytic properties, making them promising broad‐spectrum agents potential. This study concludes Boc‐protected phenylalanine‐ tryptophan‐based dipeptides can self‐assemble be used as agents. self‐assembly these offers a versatile platform for designing biomaterials tailored properties functionalities.
