Abstract Background Classical monocytes, pivotal in the innate immune response, wield considerable influence over cancer prognosis and response to immunotherapy. Our aim was construct lung adenocarcinoma (LUAD) prognostic signature associated with monocytes. Methods This study encompassed 1,822 samples drawn from diverse public datasets. Initially, we conducted an comprehensive analysis of single-cell RNA sequencing data obtained Gene Expression Omnibus (GEO), identifying 145 marker genes specific classical monocyte. Univariate Cox regression Least Absolute Shrinkage Operator (LASSO) analyses were performed on TCGA training cohort a for monocyte genes. Result Patients LUAD categorized into high- low-risk groups based polygenic derived these The predictive capacity this robustly validated across distinct clinical subgroups GEO cohorts. Notably, emerged as independent indicator via multivariate analysis. Low-risk patients exhibited heightened cell infiltration, correlating inversely inflammatory activity risk score. Conversely, group demonstrated amplified abundance diversity T-cell receptor (TCR) repertoires, higher Immunophenotype scores (IPS), lower Tumor Immune Dysfunction Exclusion (TIDE) scores. Significantly, assessment three immunotherapy cohorts showcased superior immunotherapeutic among compared their high-risk counterparts. Conclusion In summary, our introduces novel rooted that proficiently prognosticates predicts patients. Supplementary Information: information is uploaded attached document.

Cancer cells generally exhibit 'iron addiction' phenotypes, which contribute to their vulnerability ferroptosis inducers. Ferroptosis is a newly discovered form of programmed cell death caused by iron-dependent lipid peroxidation. In the present study, pacidusin B, dichapetalin-type triterpenoid from Phyllanthus acidus (L.) Skeels (Euphorbiaceae), induces in HT1080 human fibrosarcoma line. Cells treated with B exhibited morphological characteristic 'ballooning' phenotype ferroptosis. The biochemical hallmarks were also observed B-treated cells. Both oxidative stress and ER play significant roles B-induced activation PERK-Nrf2-HO-1 signaling pathway led iron overload, while inhibition GPX4 further sensitized cancer Furthermore, molecular docking study showed that docked same pocket xCT as inducer erastin. These results revealed exerts anticancer effects via inducing ER-mediated ferroptotic death.

To integrate machine learning and multiomic data on lactylation-related genes (LRGs) for molecular typing prognosis prediction in lung adenocarcinoma (LUAD). LRG mRNA long non-coding RNA transcriptomes, epigenetic methylation data, somatic mutation from The Cancer Genome Atlas LUAD cohort were analyzed to identify lactylation cancer subtypes (CSs) using 10 multiomics ensemble clustering techniques. findings then validated the GSE31210 GSE13213 cohorts. A model was developed identified hub LRGs divide patients into high- low-risk groups. effectiveness of this validated. Up to now, accurate stable predictive prognostic indicator for revealing progress novel strategies of LUAD is infrequent, nonetheless. Diversified programmed cell death (PCD) has been widely confirmed that participated occurrence development various malignant tumors, respectively. In this research, we integrated fourteen types PCD, bulk multi-omic data TCGA-LUAD other cohorts gene expression omnibus (GEO) clinical patients develop our analysis. Low mRNAsi LUAD predicted a better prognosis. Eight genes ( GNG7, EIF5A, ANLN, FKBP4, GAPDH, GNPNAT1, E2F7, CISH ) associated with were screened to establish risk model. The differentially expressed between high and low groups mainly enriched metabolism, cell cycle functions pathway. score group had higher immune scores. Patients lower TIDE scores immunotherapy outcomes. In addition, was effective assessing drug sensitivity LUAD. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) data showed that eight lines, knockdown EIF5A reduced invasion migration ability cells. Unfortunately, diagnostic biomarkers currently lacking, limiting options clinic. To discover proteins that have potential for biomarker applications, we performed an in-depth quantitative proteomic analysis on a cohort Filipino NSCLC adenocarcinoma (LUAD) Differentially expressed (DEPs) were obtained by using tandem mass tag (TMT) labeling and spectrometry (MS)-based proteomics. A total 6240 quantified identified with 3155 significantly upregulated 1248 downregulated. Integration result curated transcriptome data allowed identification 33 potential.
