Abstract Background Porcine hepatitis E is a zoonotic infectious disease caused by swine virus (HEV), open reading frames 3 an important virulence protein of porcine HEV, which plays role in the release viral particles and host innate immune response, regulation autophagy apoptosis, etc., but its main function pathogenic mechanism are not perfect current research. Results In our study, adenoviruses ADV4-ORF3 ADV4-GFP were successfully constructed mediated overexpression enhanced green fluorescent (EGFP)-ORF3 EGFP HepG2 cells. A total 217 differentially expressed messenger RNAs (mRNAs) screened high-throughput sequencing, 27 statistically significant genes for further quantitative real-time reverse transcription (qRT-PCR) verification functional enrichment (Gene Ontology [GO] Kyoto Encyclopedia Genes Genomes [KEGG]). They mainly involved 6 pathways: cellular response to unfolded protein, Inflammatory cytokine activity, TNF signaling pathway, Influenza A, Pathways cancer. Background The relationship between GPR and hepatitis E has not been reported. This study evaluates the levels in AHE patients HEV-ALF explores role of prognosis patients, offering new strategies methods for diagnosis patients. Methods Serum samples were collected from 206 217 admitted to First Affiliated Hospital Zhejiang University School Medicine, Suzhou Yongding Hospital, Nanjing Medical January 1, 2017, November 30, 2024. Results level group was significantly higher than ( p < 0.001). OPLS-DA analysis revealed that INR, TBIL, PLT, GPR, TCH, ALT, AFP, TP, ALB, AST major influential factors occurrence HEV-ALF. showed good diagnostic performance with an AUC 0.701. DCA further supported utility across a range threshold probabilities. Regarding prediction 30-day mortality, non-survival compared survival highlighted as most factor predicting mortality. demonstrated 0.703. results also indicated GPR’s strong decision-making ability mortality Conclusion is highly expressed closely related their prognosis.
