Abstract The rising incidence rate of depression presents a substantial threat to human well‐being. Semen Trigonellae (ST), the dried mature seeds Trigonella foenum‐graecum L., has long‐standing traditional reputation for alleviating anxiety and hopelessness. However, anti‐depressant mechanism ST remains poorly understood. This study aimed assess ST's anti‐depressant, as well explore its potential from gut microbial aspect. Kunming mice were challenged by lipopolysaccharide (LPS) induce depression‐like behavior then orally administrated with aqueous extract ST. behavioral test, hippocampal serum biochemical indicators detected effect We utilized full‐length 16S rRNA gene sequencing investigate how influences microbiota modulation. Administration mitigated LPS‐induced behaviors. reversed decrease in 5‐hydroxytryptamine (5‐HT) brain‐derived neurotrophic factor (BDNF) levels while reducing tumor necrosis α (TNF‐α), interleukin 1β (IL‐1β), 6 (IL‐6). Microbial analysis revealed that administration markedly shifted structure, dramatically dose‐dependently increased abundance Ligilactobacillus murinus animalis . Experimentally, oral live L LPS‐challenged yielded similar effects ameliorating depression, elevating 5‐HT BDNF, proinflammatory cytokines.
