Abstract Abelmoschus manihot is an important medicinal and edible plant known for its functional secondary metabolites. However, little about the key genes involved in production of metabolites A. manihot. This largely due to a lack effective gene expression detection systems manihot, thus screening real-time PCR reference prerequisite. In this study, 11 candidate were screened cloned from their stability was evaluated different tissues using four algorithms: geNorm, NormFinder, BestKeeper, RefFinder. The eIF PP2A1 highest, while that tubulin alpha (TUA) lowest. combined use two most stable genes, PP2A1, met experimental requirements normalizing Furthermore, transcription factors bHLH147 bHLH148 further validated by data normalization. study identified potential manihot tissues, paving way analysis dissecting metabolite regulation mechanisms

Abstract Skin, as the first line of defence human body, is exposed to dangers such overheating substances, ultraviolet rays, and environmental pollutants, incidence skin diseases increasing annually. Oxidative stress plays a dominant role in most diseases. Abelmoschus manihot (L.) medic flower (TEA) traditional Chinese medicine widely used treat injuries water fire scalds. It has been reported that TEA excellent antioxidant effects. In this study, we aimed explore mitochondrial protection effects H 2 O -mediated HaCaT cell damage. cells were incubated with simulate oxidative skin. The effect on was also evaluated. Cell morphology observed via inverted microscopy, viability measured MTT reagent. stained Hoechst 33,324 solution. Reactive oxygen species (ROS), superoxide dismutase (SOD), malondialdehyde (MDA) ATP detection kits detect corresponding indicators. membrane potential detected by JC-1. RT-PCR mRNA mtDNA expression. expression target protein Western blotting immunofluorescence. triggered damage cells, which manifested apoptosis, increased ROS MDA contents, decreased SOD activity. activates KEAP1/Nrf2/NQO1 signalling pathway, decreases intracellular KEAP1 slightly increases Nrf2 NQO1 proteins, further causing stress, resulting changes potential, reduction copy number, PGC-1α TFAM proteins. addition respiratory chain genes proteins decreased. promoted activated downstream response, alleviated caused . ML385 an inhibitor, under protective are inhibited. When knocked down, mitochondria protects from -induced through KEAP1/Nrf2/NQO1/PGC-1α/TFAM pathway.

Carboxymethyl tamarind xyloglucan (CMTXG) was synthesized by with sodium chloroacetate in 50% ethanol. The effects of carboxymethylation on the structure and properties were studied, its application potential cosmetics evaluated. results showed that increased molecular weight, enhanced water solubility stability, decreased apparent viscosity, promoted xyloglucan's cosmetics. CMTXG moderate substitution (CMT-M) exhibited superior hygroscopic moisturizing effects, indicating does not need to pursue high degree substitution. rate CMT-M 81.08% after 12 h, surpassing hyaluronate (55.79%) glycerin (59.52%). antioxidant antibacterial activity modification. Toxicity assessment indicated can improve long-term tolerance larvae zebrafish. Furthermore, cream demonstrated a effect compared commercial creams. research findings are expected provide an effective bioactive moisturizer candidate for daily chemical industry.
