Alzheimer's disease (AD) is a chronic neurodegenerative condition characterized by progressive cognitive impairment. While the formation of β-amyloid plaques and neurofibrillary tangles are hallmarks features AD, downstream consequence these byproducts disruption cholinergic glutamatergic neural systems. Growing evidence for existence interplay between AChE NMDARs has opened up new venues discovery novel ligands endowed with anticholinesterase NMDAR-blocking activity. Plants belonging to stachys genus have been extensively explored having broad range therapeutic applications used traditionally millennia, treat various CNS-related disorders, which makes them ideal source therapeutics. The present study was designed identify natural dual-target inhibitors NMDAR deriving from their potential use in AD. Using molecular docking, drug-likeness-profiling, MD simulation MMGBSA calculations, an in-house database biomolecules pertaining shortlisted based on binding affinity, overall stability critical ADMET parameters. Pre- post-MD analysis revealed that Isoorientin effectively binds vital interactions, exhibits stable behavior minor fluctuations relative two clinical drugs as positive control, displays strong consistent interactions lasted majority simulation. Findings this elucidated rationale behind traditional Stachys plants treatment AD could provide impetus development therapeutics treatment.Communicated Ramaswamy H. Sarma

The increase in people’s longevity has, consequently, led to more brain involvement and neurodegenerative diseases, which can become complicated lead chronic degenerative thereby presenting greater public health problems. Medicinal plants have been used since ancient times contain high concentrations of molecules, including polyphenols. It has proven that polyphenols, are present various natural sources provide curative effects against diseases disorders through neuroprotective effects. These mainly attributed their ability cross the blood–brain barrier, eliminate reactive oxygen species, cause chelation metal ions. Polyphenols concentration neurotrophic factors bind directly membrane receptors these factors, modulate activate signaling cascades allow plasticity, survival, proliferation, growth neuronal cells, allowing for better learning, memory, cognition. Moreover, polyphenols no serious adverse side resulting from consumption.

Cardiovascular disease (CVD) is a major contributor to increasing morbidity and mortality worldwide seriously threatens human health life. Cardiomyocyte death considered the pathological basis of various CVDs, including myocardial infarction, heart failure, aortic dissection. Multiple mechanisms, such as ferroptosis, necrosis, apoptosis, contribute cardiomyocyte death. Among them, ferroptosis an iron-dependent form programmed cell that plays vital role in physiological processes, from development aging immunity CVD. The dysregulation has been shown be closely associated with CVD progression, yet its underlying mechanisms are still not fully understood. In recent years, growing amount evidence suggests non-coding RNAs (ncRNAs), particularly microRNAs, long RNAs, circular involved regulation thus affecting progression. Some ncRNAs also exhibit potential value biomarker and/or therapeutic target for patients this review, we systematically summarize findings on their We focus clinical applications diagnostic prognostic biomarkers well targets treatment. Antioxidants are widely recognized for their potential health benefits, including impact on cognitive function and gut microbiome modulation. Understanding these effects is essential exploring broader clinical applications. This review aims to evaluate the of antioxidants function, with a focus findings from randomized controlled trials (RCTs). The studies involved human participants across range age groups, interventions encompassing natural antioxidant sources, such as berries, well specific vitamins. An extensive search PubMed, SCOPUS, Web Science databases identified six relevant RCTs, each evaluated bias. These focused variety antioxidant-rich products, both naturally derived sources supplemental forms. Antioxidants, vitamins C, B2, D, along polyphenols xanthohumol, fermented papaya, peanuts, berry extracts, demonstrate support promote through mechanisms that modulate diversity reduce inflammation. However, observed changes in were modest inconsistent studies. While preliminary evidence suggests may benefit heterogeneity existing limits immediate applicability. Additionally, more robust RCTs needed substantiate guide future interventions.

The neurodegenerative condition known as Parkinson’s disease (PD) is brought on by the depletion of dopaminergic neurons in basal ganglia, which brain region that controls body movement. PD occurs due to many factors, from one acknowledged effects oxidative stress pathogenic pathways play a role development disease. Antioxidants, including flavonoids, vitamins E and C, polyphenolic substances, help reduce free radicals. Consequently, this lowers risk disorders long term. Although there currently no cure for illnesses, these conditions can be controlled. treatment lessens its symptoms, helps preserve patient’s quality life. Therefore, use naturally occurring antioxidants, such polyphenols, may obtained through food or nutritional supplements have variety positive effects, has emerged an appealing alternative management strategy. Neurodegenerative diseases encompass a spectrum of disorders marked by the progressive degeneration structure and function nervous system. These conditions, including Parkinson's disease (PD), Alzheimer's (AD), Huntington's (HD), Amyotrophic lateral sclerosis (ALS), Multiple (MS), often lead to severe cognitive motor deficits. A critical component neurodegenerative pathologies is imbalance between pro-oxidant antioxidant mechanisms, culminating in oxidative stress. The brain's high oxygen consumption lipid-rich environment make it particularly vulnerable damage. Pro-oxidants such as reactive nitrogen species (RNS) (ROS) are continuously generated during normal metabolism, counteracted enzymatic non-enzymatic defenses.
