Oxidative stress plays a critical role in various physiological and pathological processes, particularly during pregnancy, where it can significantly affect maternal fetal health. In the context of viral infections, such as those caused by Human Immunodeficiency Virus (HIV) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), oxidative may exacerbate complications disrupting cellular function immune responses. Antiviral drugs, while essential managing these also contribute to stress, potentially impacting both mother developing fetus. Understanding mechanisms which antivirals examination pharmacokinetic changes pregnancy that influence drug metabolism is essential. Some research indicates antiretroviral drugs induce mitochondrial dysfunction other studies suggest their use generally safe. Therefore, concerns about long-term health effects persist. This review delves into complex interplay between antioxidant defenses, antiviral therapies, focusing on strategies mitigate potential damage. By addressing gaps our understanding, we highlight importance balancing efficacy with risks stress. High levels of oxidant stress in the form reactive species are prevalent circulation and tissues various types cardiovascular disease including heart failure, hypertension, peripheral arterial disease, stroke. Here we review role nuclear factor erythroid 2-related 2 (Nrf2), an important widespread antioxidant anti-inflammatory transcription that may contribute to pathogenesis maintenance diseases. We studies showing downregulation Nrf2 exacerbates autonomic function. Finally, discuss potential for using modulation as a therapeutic strategy diseases dysfunction.

ABSTRACT Background Several respiratory viruses, including Severe Acute Respiratory Syndrome‐Coronavirus‐2 (SARS‐CoV‐2), suppress nuclear factor‐E2‐related factor‐2 (NRF2) antioxidant response, generating oxidative stress conditions to its advantage. NRF2 has also been reported regulate the innate immune response through inhibition of interferon (IFN) pathway. However, modulation in younger individuals and correlation with IFN remain be elucidated. Methods The redox‐related genes expression was examined nasopharyngeal swabs from children attending pediatric hospital for SARS‐CoV‐2 molecular testing. Expression levels were analyzed by stratifying population according positivity, age, or presence symptoms. results correlated Types I III IFN‐stimulated (ISGs). Results We found that markedly diminished positive patients compared negative. Moreover, it higher IFNα2 IFNλ3, as well ISG15 ISG56. Interestingly, symptomatic anosmia/ageusia showed pronounced apurinic/apyrimidinic endonuclease1/redox factor 1 (APE1), together Type IFNs, ISG56, inflammasome component NLRP3. Conclusion indicate an interdependence between pathway IFN‐mediated during infection young subjects.

Respiratory viruses constitute a significant cause of illness and death worldwide. virus-associated injuries include oxidative stress, ferroptosis, inflammation, pyroptosis, apoptosis, fibrosis, autoimmunity, vascular injury. Several studies have demonstrated the involvement nuclear factor erythroid 2-related 2 (Nrf2) in pathophysiology viral infection associated complications. It has thus emerged as pivotal player cellular defense mechanisms against such damage. Here, we discuss impact Nrf2 activation on airway induced by respiratory viruses, including coronaviruses, rhinoviruses, syncytial viruses. The inhibition or deregulation pathway induces tissue damage presence infections. In contrast, demonstrates protection organ injuries. Clinical trials involving agonists are needed to define effect therapeutics tissues organs damaged

It is well established that Nrf2 plays a crucial role in anti-oxidant and anti-inflammatory functions. However, its antiviral capabilities remain less explored. Despite this, several activators have demonstrated anti-SARS-CoV-2 properties, though the mechanisms behind these effects are not fully understood. In this study, using two mouse models of SARS-CoV-2 infection, we observed absence significantly increased viral load altered inflammatory responses. Additionally, evaluated five modulators. Notably, epigallocatechin gallate (EGCG), sulforaphane (SFN), dimethyl fumarate (DMF) exhibited significant effects, with SFN being most effective. did impact entry but appeared to inhibit main protease (M

Viral respiratory infections (VRIs) are a leading cause of morbidity and mortality worldwide, making them significant public health concern. During infection, viruses, including Influenza virus, SARS-CoV-2, syncytial virus (RSV), trigger an antiviral immune response, specifically boosting the inflammatory response that plays critical role in their pathogenesis. The induced by viruses can be double-edged sword since it initially to protective/reparative from virus-induced injuries. Still, also detrimental host cells tissues. However, mechanisms differentiate complex crosstalk between favorable responses harmful poorly understood. Recent reports of fatal meningoencephalitis from Kerala, South India, are creating ripples among medical and health administrations. Free-living amoebae (FLA) have been attributed to these infections. Despite their free-living nature, recognized as potential opportunistic organisms that occasionally cause infections in immunocompetent immunocompromised individuals. The pathophysiology, epidemiology, clinical course diseases caused by FLA not completely understood. Exposure humans animals is imminent given presence the air, water, soil. Human infections, although infrequent, can lead severe morbidity mortality evidenced recently emerging India. Since were noticed children, it unlikely they immunocompromised. Therefore, this editorial, we speculate on possibility frequent upper respiratory tract viral predisposing factors for infection with FLA.
