In
2022,
the
number
of
mpox
cases
spiked
worldwide,
leading
to
a
surge
in
scientific
research
on
members
Orthopoxvirus
genus
and
discovery
new
compounds
exhibiting
anti-orthopoxvirus
activity.
This
work
is
devoted
synthesis
containing
an
adamantane
fragment
evaluation
their
activity
against
vaccinia
virus,
offering
possible
mechanism
antiviral
action

agents.

Recent
emergence
of
zoonotic
monkeypox
virus
(Mpox)
in
human
has
triggered
the
virologists
to
develop
plausible
preventive
measures.
Hitherto,
our
understanding
on
mechanism
immunopathogenesis
Mpox
infection
is
elusive.
However,
available
experimental
evidences
suggest
induction
inflammation
as
main
cause
pathogenesis.
Toll-like
receptors
(TLRs)
are
critical
initiating
and
modulating
host
immune
response
pathogens.
Inflammatory
responses
observed
various
poxvirus
infections
have,
fact,
been
shown
be
mediated
through
TLR
activation.
Therefore,
by
silico
approaches,
this
study
seeks
identify
antigen(s)
(MAg)
that
most
likely
interact
with
cell-surface
TLRs.
The
proteomics
data
UniProt
database
contain
174
protein
sequences,
among
which
105
immunoreactive
proteins
were
modeled
for
3D
structure
examined
comparative
protein-protein
interactions
TLRs
molecular
docking
dynamics
simulation.
F14,
an
8.28
kDa
infective
Mpox,
was
found
exhibit
strong
binding
affinity
(ΔG=-12.5
Kcal
mol-1)
TLR1/2

This
study
presents
the
development
and
validation
of
a
genomic
surveillance
strategy
using
Whole
Genome
Sequencing
(WGS)
on
normalized
pooled
samples
to
detect
monitor
SARS-CoV-2
variants.
A
bioinformatics
pipeline
was
designed
specifically
for
analyzing
WGS
data
validated
simulated
datasets,
reference
materials,
clinical
collected
during
key
periods
Delta
Omicron
variant
emergence.
The
approach
evaluated
its
accuracy
in
estimating
abundance
at
both
Phylogenetic
Assignment
Named
Global
Outbreak
(PANGO)
lineage
level
World
Health
Organization
(WHO)
level.
From
simulation
method
achieved
an
overall
sensitivity
99.1%
positive
predictive
value
(PPV)
99.9%
detecting
variants
WHO
At
PANGO
level,
it
82.8%
PPV
77.4%
when
predicted
considered
accurate
if
shared
more
than
90%
markers
with
any
true
present
sample.
