In the 21st century, thanks to advances in biotechnology and developing pharmaceutical technology, significant progress is being made effective drug design. Drug targeting aims ensure that acts only pathological area; it defined as ability accumulate selectively quantitatively target tissue or organ, regardless of chemical structure active substance method administration. With targeting, conventional, biotechnological gene-derived drugs body’s organs, tissues, cells can be transported specific regions. These systems serve carriers regulate timing release. Despite having many advantageous features, these have limitations thoroughly treating complex diseases such cancer. Therefore, combining with nanoparticle technologies imperative treat cancer at both local systemic levels effectively. The nanocarrier-based delivery involves encapsulating target-specific molecules into polymeric vesicular systems. Various (DDS) were investigated discussed this review article. first part passive systems, hydrogels, thermoplastics, microdevices transdermal-based second carrier nanobiotechnology (carbon nanotubes, nanoparticles, coated, pegylated, solid lipid nanoparticles smart nanogels). third part, advantages discussed, finally, market research commercial used nanotechnological approaches was included.

Plasmalogens (vinyl-ether phospholipids) are an emergent class of lipid drugs against various diseases involving neuro-inflammation, oxidative stress, mitochondrial dysfunction, and altered metabolism. They can activate neurotrophic neuroprotective signaling pathways but low bioavailabilities limit their efficiency in curing neurodegeneration. Here, liquid crystalline nanoparticles (LNPs) created for the protection non-invasive intranasal delivery purified scallop-derived plasmalogens. The vivo results with a transgenic mouse Parkinson's disease (PD) model (characterized by motor impairments α-synuclein deposition) demonstrate crucial importance LNP composition, which determines self-assembled nanostructure type. Vesicle hexosome nanostructures small-angle X-ray scattering) display different efficacy nanomedicine-mediated recovery function, balance, transcriptional regulation (e.g., reduced neuro-inflammation PD pathogenic gene expression). Intranasal vesicular hexosomal plasmalogen-based treatment leads to improvement behavioral symptoms downregulation Il6, Il33, Tnfa genes. Moreover, RNA-sequencing lipidomic analyses establish dramatic effect nanomedicines on amelioration, metabolism, type number responsive transcripts that may be implicated neuroregeneration.

Parkinson’s disease (PD) and other chronic debilitating neurodegenerative diseases (NDs) impose a substantial medical, emotional, financial burden on individuals society. The origin of PD is unknown due to complex combination hereditary environmental risk factors. However, over the last several decades, significant amount available data from clinical experimental studies has implicated neuroinflammation, oxidative stress, dysregulated protein degradation, mitochondrial dysfunction as primary causes neurodegeneration. new gene-editing techniques hold great promise for research therapy NDs, such PD, which there are currently no effective disease-modifying treatments. As result, gene may offer treatment options, transforming our ability treat this disease. We present detailed overview novel delivery vehicles, essential their successful implementation in both cutting-edge prospective therapeutics. Moreover, we review most recent advancements CRISPR-based applications therapies better understanding treating PD. explore benefits drawbacks using them range brain, emphasizing some fascinating possibilities.
