Phloretin and its derivatives are dihydrochalcone compounds with diverse pharmacological properties biological activities, offering significant potential for applications in the food pharmaceutical industries. Resveratrol, a polyphenolic compound known for its diverse biological activities, has demonstrated multiple pharmacological effects, including anti-inflammatory, anti-aging, anti-diabetic, anti-cancer, and cardiovascular protective properties. Recent studies suggest that these effects are partly mediated through the regulation of macrophage polarization, wherein macrophages differentiate into pro-inflammatory M1 or anti-inflammatory M2 phenotypes. Our review highlights how resveratrol modulates polarization various signaling pathways to achieve therapeutic effects. For example, can activate senescence-associated secretory phenotype (SASP) pathway inhibit signal transducer activator transcription (STAT3) sphingosine-1-phosphate (S1P)-YAP axes, promoting suppressing thereby inhibiting tumor growth. Conversely, it promote suppress by NF-κB activating PI3K/Akt AMP-activated protein kinase (AMPK) pathways, thus alleviating inflammatory responses. Notably, effect on is concentration-dependent; moderate concentrations tend while higher may favor polarization. Background/Objectives: Cutibacterium acnes (C. acnes), a bacterium residing in hair follicles, triggers acne by inducing monocyte-mediated inflammatory cytokine production. Gedunin, limonoid derived from Azadirachta indica (commonly known as neem), is renowned for its antifungal, antimalarial, anticancer, anti-inflammatory, and neuroprotective effects. However, role mitigating C. acnes-induced skin inflammation remains unexplored. This study investigates the anti-inflammatory effects of gedunin on elucidates underlying mechanisms. Methods: The activity was assessed using RAW 264.7 mouse macrophage cells bone-marrow-derived macrophages (BMDMs). Key mediators, including interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin-6 (IL-6), were evaluated. Mechanistic studies focused nuclear factor-kappa B (NF-κB) mitogen-activated protein kinase (MAPK) signaling pathways, along with NOD-like receptor pyrin domain-containing 3 (NLRP3) inflammasome. An vivo model employed to examine gedunin's therapeutic efficacy. Results: Gedunin significantly reduced expression IL-1β, TNF-α, iNOS, COX-2, IL-6 cells. Melinjo (Gnetum gnemon L.) seeds are rich in resveratrol (RES) and its dimers, gnetin C gnemonoside A. They have various pharmacological benefits, antioxidant anti-inflammatory properties; however, their pharmacokinetics, especially animal models, must be better understood. This study aimed to elucidate the pharmacokinetics of melinjo seed extract (MSE) components rats. Plasma samples were collected up 24 h after intergastric administration MSE (1000 mg/kg), C, or A (51 μmol/kg each). RES was present as conjugates, with maximum plasma concentration (1065 nM) achieved at 6 postadministration MSE. The concentrations following sustained, absorption rates, based on AUC–dose relationship, reached 13.7 4.8%, respectively, higher than that each standard alone. These findings indicate enhances derivatives, potentially supporting therapeutic applications functional foods.

The aim of this study was to structurally characterize resveratrol-piperazine cocrystal forms using the single-crystal X-ray diffraction technique. Osteoporosis is a prevalent metabolic bone disorder characterized by decreased mineral density and increased fracture risk, particularly among aging populations. While conventional pharmacological treatments exist, they often have adverse effects, necessitating the search for alternative therapies. Resveratrol, naturally occurring polyphenol, has gained significant attention its potential osteoprotective properties through various molecular mechanisms. This systematic review aims to comprehensively analyze pathways which resveratrol protects against osteoporosis. Using an advanced strategy in Scopus, PubMed, Web of Science databases, we identified 513 potentially relevant articles. After title abstract screening, followed full-text review, 28 studies met inclusion criteria. The selected comprised 14 vitro studies, 8 mixed vivo 6 1 cross-sectional study postmenopausal women. Our findings indicate that exerts effects enhancing osteoblast differentiation activation Phosphoinositide 3-Kinase/Protein Kinase B (PI3K/Akt), Sirtuin (SIRT1), AMP-Activated Protein (AMPK), GATA Binding (GATA-1) while simultaneously inhibiting osteoclastogenesis suppressing Mitogen-Activated (MAPK) TNF Receptor-Associated Factor 6/Transforming Growth Factor-β-Activated (TRAF6/TAK1).
