Abstract Successful infection strategies must balance pathogen amplification and persistence. In Toxoplasma gondii , this is accomplished through differentiation into dedicated cyst-forming chronic stages that avoid clearance by the host immune system. The transcription factor BFD1 both necessary sufficient for stage conversion; however, its regulation not understood. We examine five factors transcriptionally activated BFD1. One of these a cytosolic RNA-binding protein CCCH-type zinc finger family, which we name BFD2. Parasites lacking BFD2 fail to induce are consequently unable fully differentiate in culture or mice. interacts with transcript stress-dependent manner. Deletion reduces levels, but mRNA abundance. reciprocal effects on translation outline positive feedback loop enforces commitment differentiation. helps explain how parasites commit gene-expression program elucidates between proliferation persistence achieved over course infection.

Abstract Dinoflagellates from the family Symbiodiniaceae are phototrophic marine protists that engage in symbiosis with diverse hosts. Their large and distinct genomes characterized by pervasive gene duplication large-scale retroposition events. However, little is known about role scale of horizontal transfer (HGT) evolution this algal family. In other dinoflagellates, high levels HGTs have been observed, linked to major genomic transitions, such as appearance a viral-acquired nucleoprotein originated via HGT DNA virus. Previous work showed different hosts actively infected viral groups, giant viruses ssRNA viruses, may play an important coral health. Latent infections also occur, whereby could persist cytoplasm or integrate into host genome provirus. This hypothesis received experimental support; however, cellular localization putative latent their taxonomic affiliation still unknown. addition, despite finding sequences some Symbiodiniaceae, origin, breadth, metabolic potential not explored. To address these questions, we searched for viral-derived proteins thirteen genomes. We found fifty-nine candidate gave rise twelve phylogenies across ten describe virus-related sequences, structure, context. These results lead us propose model explain origin fate acquisitions.

Abstract Background Although the end of COVID-19 as a public health emergency was declared on May 2023, still new cases infection are reported and risk remains variants emerging that may cause surges in deaths. While clinical symptoms have been rapidly defined worldwide, basic body responses pathogenetic mechanisms acting patients with SARS-CoV-2 over time until recovery or death require further investigation. The understanding molecular underlying development course disease is essential designing effective preventive therapeutic approaches, ultimately reducing mortality spreading. Methods current investigation aimed to identify key genes engaged uncover their implication severity. To achieve this goal high-throughput RNA sequencing peripheral blood samples collected from healthy donors performed. resulting sequence data were processed using wide range bioinformatics tools obtain detailed modifications within five transcriptomic phenomena: expression long non-coding RNAs, alternative splicing, allel-specific circRNA production. silico procedure completed functional analysis identified alterations. Results revealed has significant impact multiple encoding ribosomal proteins (RPs). show these differ not only terms but also manifest biases splicing ASE ratios. integrated exposed RPs mostly affected pathways processes related infection—COVID-19 NOD-like receptor signaling pathway, SARS-CoV-2-host interactions response virus. Furthermore, our results linked intronic exonic circular differentiations infection, suggesting events play crucial role mRNA maturation transcription during disease. Conclusions By elucidating genetic induced by virus, research provides information can be employed create targeted strategies for future treatment COVID-19. Moreover, findings highlight potentially promising biomarkers early assessment critically ill patients.
