ABSTRACT Evaluating SARS-CoV-2 viral loads in nasopharyngeal (NP) and saliva samples, factors affecting loads, the performance of rapid antigen testing (RAT) have not been comprehensively conducted during Omicron epidemic. This prospective study included outpatients enrolled variant period Japan. Paired NP swab samples were collected to measure by reverse transcription-quantitative polymerase chain reaction (RT-qPCR). The total testing process harmonization is central to laboratory medicine, leading the test's effectiveness. In this opinion paper five phases of TTP are analyzed, describing, and summarizing critical issues that emerged in each phase with SARS-CoV-2 serological tests have affected their Testing screening population was essential for defining seropositivity and, thus, driving public health policies management COVID-19 pandemic. SARS-CoV-2 diagnostic tests have become an important tool for pandemic control. Among the alternatives COVID-19 diagnosis, antigen rapid (Ag-RDT) are very convenient and widely used. However, as variants may continuously emerge, replacement of reagents be required to maintain sensitivity Ag-RDTs. Here, we describe development validation Ag-RDT during outbreak Omicron variant, including characterization a new monoclonal antibody (anti-DTC-N 1B3 mAb) that recognizes Nucleocapsid protein (N). The anti-DTC-N mAb recognized sequence TFPPTEPKKDKKK located at C-terminus N main concern. Accordingly, prototypes using mAB detected all variants—Wuhan, Alpha, Gamma, Delta, P2 Omicron. performance best prototype (sensitivity 95.2% samples with Ct ≤ 25; specificity 98.3% overall accuracy 85.0%) met WHO recommendations. Moreover, results from patients’ follow-up study indicated that, if performed within first three days after onset symptoms, displayed 100% sensitivity. Thus, developed herein constitute alternative tools point-of-care diagnosis epidemiological surveillance.

Abstract Purpose To investigate the perception of SARS-CoV-2 detection methods, information sources, and opinions on appropriate behavior after receiving negative or positive test results. Methods In a questionnaire-based, cross-sectional study conducted between September 1 November 17, 2021, epidemiological, behavioral, COVID-19-related data was acquired from public in Munich, Germany. Results Most 1,388 participants obtained online media (82.8%) as well state federal authorities (80.3%). 93.4% believed accuracy PCR testing 41.2% rapid antigen tests (RATs). In Korea, during the early phase of coronavirus disease 2019 (COVID-19) pandemic, we responded to uncertainty treatments under various conditions, consistently playing catch up with speed evidence updates. Therefore, there was high demand for national-level evidence-based clinical practice guidelines clinicians in a timely manner. We developed and updated living recommendations through transparent development process multidisciplinary expert collaboration.The National Evidence-based Healthcare Collaborating Agency (NECA) Korean Academy Medical Sciences (KAMS) collaborated develop trustworthy guidelines. The NECA-supported methodological sections 8 professional medical societies KAMS worked experts, 31 were involved annually. total 35 questions, including medications, respiratory/critical care, pediatric emergency diagnostic tests, radiological examinations.An search began March 2021 monthly updates performed. It expanded other areas, interval organized by steering committee owing priority changes.
