Immunoproteasome (IP) is induced by pro-inflammatory stimuli such as interferon gamma to regulate inflammation and immunity. Asthma patients with airway type 2 high (e.g., IL-13) demonstrate more eosinophils hyperresponsiveness (AHR) less gamma. The role of IP in regulating eosinophilic AHR has not been investigated. This study was aimed determine how regulates using LMP7 (a subunit IP) deficient mouse lungs, precision-cut lung slices (PCLS), cultured human epithelial cells treated IL-13 the absence or presence an inhibitor ONX-0914 exogenous IP. KO lungs had significantly IL-4Rα protein expression than wildtype (WT) mice. Following treatment PCLS, mice contraction WT mice, which coupled increased eotaxin-2 levels. inhibition resulted eotaxin-3 release. PCLS AHR. Collectively, these data demonstrated that promotes degradation IL-4Rα, while inhibits Enhancement activity may serve alternative approach reduce severity
