Abstract Background Ninjin’yoeito (NYT), a traditional Japanese Kampo medicine, improves the depression and anxiety in humans animals, rendering it novel therapeutic option for post-stroke (PSD). Furthermore, physical exercise is an important nonpharmacological therapy major depressive disorder. The components of NYT or exert antidepressant effects through increased expression neurotrophic factors reduced neuroinflammation brain. However, mechanisms underlying PSD remain unclear. Therefore, we examined rat model PSD. Methods Rats were divided into five groups: PSD, with NYT, (Ex), (NYT + Ex), control (sham). was induced by microinjection endothelin-1 left medial prefrontal cortex chronic unpredictable mild stress 3 days per week. A diet containing 3% administered to rats one day after stroke induction. Exercise conducted using motorized treadmill three week, starting stroke. interventions lasted four weeks. Serum corticosterone levels, depression-like behavior, hippocampal pathophysiology, including brain-derived factor (BDNF), precursor BDNF (proBDNF), doublecortin (DCX), NeuN, glial cell activation, tumor necrosis factor-α (TNF-α), examined.
