Viral infectious diseases cause millions of deaths and severe illness all around the world affecting public health economics. vaccines are helping to fight against viral diseases, but current expensive unavailable, especially in poor developing countries. When measuring entire manufacturing processing, downstream processing is major cost production. Our goal for this research develop a low-cost alternative platform new vaccine infrastructures. We have developed novel osmolyte flocculation method particles. To create purification several types particles, we used two model viruses: porcine parvovirus (PPV) Sindbis virus (SINV). PPV non-enveloped virus, one smallest known mammalian viruses with diameter approximately 20 nm. The enveloped SINV, has size 48-52 Using mannitol demonstrated recovery both by diafiltration using micropore membrane. This will allow easy scale-up production scale creates platform. lab’s previous study showed that was specific as compared proteins which present contaminants process. preferential due active hydrophobic surface differences on protein surfaces. studied effect membrane pore were able achieve 60% 0.1 μm 500 kDa filters. Recovery SINV 79% 96% filter. Increasing concentration results enhanced at high concentration, pores can get blocked, causing fouling. also examined purity recovered samples DNA contaminants. In conclusion, process purify recover particles large filters, decrease overall costs.
