Abstract Rationale Candidozyma auris (formerly Candida ) is a globally emerging potentially multi-drug resistant human pathogenic yeast. Biofilms are the predominant lifeforms of microorganisms, contributing to over 80% infections, including those caused by Candida species like C. albicans, parapsilosis and Candidozyma auris. Abstract Resistance to echinocandins in the recently emerged pathogen Candidozyma auris (previously known as Candida ) poses significant clinical challenges. This study reports vitro evolution of a caspofungin-sensitive C. clade II isolate acquire caspofungin resistance. We sequenced whole genome parental strain and its drug-resistant progeny. The resistant isolates harboured mutations FKS1 gene altering conserved residue (S639) also mutated caspofungin-resistant isolates; demonstrating that FKS1-S639 are sufficient confer resistance caspofungin. Using these strains, we examined cross-resistance & collateral sensitivity other antifungal drugs cell wall stress, ultrastructure, biofilm formation, virulence. ABSTRACT Background Epidemiological surveillance of Candidozyma sp. has become important because many species this new genus have been reported to be responsible for nosocomial outbreaks and exhibit elevated minimal inhibitory concentrations (MIC) one or more classes antifungal drugs. With rising multidrug-resistant yeast pathogens, conventional antifungals are becoming less effective, urging the need for adjuvants that enhance their activity at lower doses. This study evaluated synergistic of antimicrobial peptidomimetics (TM8 and RK758) or colistin sulphate in combination with against Candida albicans, C. tropicalis, parapsilosis, Meyerozyma guilliermondii, Nakaseomyces glabratus, Pichia kudriavzevii Kluyveromyces marxianus, Candidozyma auris using checkerboard microdilution test. RK758 was fluconazole 78% isolates, remaining 22% isolates still showing partial synergy; it showed synergy amphotericin B 56% caspofungin, exhibited either synergy. TM8 44% (with another 44%) 67% caspofungin isolates. Colistin No antagonism observed any combinations. Additionally, a time-kill assay further demonstrated between RK758. This study included 24 isolates of the Candidozyma haemuli species complex from patients in Brazilian Amazon between 2021 and 2024. These were identified by sequencing as C. duobushaemuli (54.2%), sensu stricto (29.2%), var. vulneris (12.5%), pseudohaemuli (4.2%). The finding represents first case reported Brazil. Haplotype phylogenetic analysis these species, along with other Brazil, revealed low intraspecific genetic diversity. Resistance to at least one antifungal was observed 83.3% isolates, multidrug resistance 58.3%, including isolate resistant all tested antifungals. demonstrated active biofilm production, lytic enzyme activity, thermotolerance.
