Light exerts a wide range of effects on mammalian physiology and behaviour. As well as synchronising circadian rhythms to the external environment, light has been shown modulate autonomic neuroendocrine responses regulating sleep influencing cognitive processes such attention, arousal performance. The last two decades have seen major advances in our understanding retinal photoreceptors that mediate these non-image forming light, neural pathways molecular mechanisms by which are generated entrained light/dark (LD) cycle. By contrast, lighting influences more equivocal. different complex. direct indirect may also occur due disrupted entrainment. Despite widespread use LD cycles study role cognition, experimental protocols used subtly function not comparable. Moreover, will disrupt alter physiological arousal, both known cognition. Studies behavioural assays dependent upon sensory processes, contribute their variable findings. Here we propose studies addressing conditions must account for rhythms, if fully understand basis effects.

Organisms adapt to day-night cycles through highly specialized circadian machinery, whose molecular components anticipate and drive changes in organism behavior metabolism. Although many effectors of the immune system are known follow daily oscillations, role clock response acute infections is not understood. Here we show that modulates inflammatory during infection with pathogen Salmonella enterica serovar Typhimurium (S. Typhimurium). Mice infected S. were colonized higher levels developed a proinflammatory early rest period for mice, compared other times day. We also demonstrate functional required optimal colonization maximal induction several genes. These findings point clock-regulated mechanism activation against an enteric may suggest potential therapeutic strategies chronopharmacologic interventions.

Circadian disruption is pervasive and can occur at multiple organizational levels, contributing to poor health outcomes individual population levels. Evidence points a bidirectional relationship, in that circadian increases disease severity many diseases disrupt rhythms. Importantly, increase the risk for expression development of neurologic, psychiatric, cardiometabolic, immune disorders. Thus, harnessing rich findings from preclinical translational research biology enhance via circadian-based approaches represents unique opportunity personalized/precision medicine overall societal well-being. In this Review, we discuss implications human using bench-to-bedside approach. science applied clinical population-based Given broad regulation health, Review focuses its discussion on selected examples metabolic, cardiovascular, allergic, immunologic disorders highlight interrelatedness between potential interventions, such as bright light therapy exogenous melatonin, well chronotherapy improve and/or modify outcomes.
