This research aimed to explore the healing impacts of Melittin treatment on gastrocnemius muscle wasting caused by immobilization with a cast in rabbits. Twenty-four rabbits were randomly allocated four groups. The procedures included different injections: 0.2 mL normal saline Group 1 (G1-NS); 4 μg/kg 2 (G2-4 Melittin); 20 3 (G3-20 and 100 (G4-100 Melittin). Ultrasound was used guide injections into rabbits’ atrophied calf muscles following two weeks via casting. Clinical measurements, including length calf, compound action potential (CMAP) tibial nerve, thickness, assessed. Additionally, cross-sectional slices fibers examined, immunohistochemistry Western blot analyses performed therapy. mean regenerative changes, as indicated clinical parameters, significantly more pronounced than other groups (p < 0.05). Furthermore, area immunohistochemical indicators exceeded those remaining analysis also showed significant presence anti-inflammatory angiogenic cytokines compared others therapy at higher dosage can efficiently activate regeneration lower doses or saline.

The pathogenesis of membranous nephropathy (MN) involves podocyte injury that is attributed to inflammatory responses induced by local immune deposits. Astragaloside IV (AS-IV) known for its robust anti-inflammatory properties. Here, we investigated the effects AS-IV on passive Heymann nephritis (PHN) rats and TNF-α-induced podocytes determine underlying molecular mechanisms MN. Serum biochemical parameters, 24-h urine protein excretion renal histopathology were evaluated in PHN control rats. expression tumor necrosis factor receptor associated 6 (TRAF6), phosphorylation nuclear kappa B (p-NF-κB), proinflammatory cytokines (TNF-α, IL-6 IL-1β) ubiquitination TRAF6 measured podocytes. We detected a marked increase mRNA TNF-α, IL-1β abundance p-NF-κB within tissues Conversely, there was reduction K48-linked TRAF6. Additionally, effective ameliorating serum creatinine, proteinuria, This effect concomitant with suppression NF-κB pathway activation decreased IL-6, levels promoting ubiquitin conjugation TRAF6, which triggered ubiquitin-mediated degradation. In summary, averted impairment podocytes, likely modulating response through TRAF6/NF-κB axis. Targeting holds therapeutic promise managing
