Insufficient sleep is becoming increasingly common and contributes to many health issues. To combat sleepiness, caffeine consumed daily worldwide. Thus, consumption restriction often occur in succession. The gut microbiome can be rapidly affected by either one’s status or intake, whereas the synergistic effects of a persistent caffeine-induced remain unclear. In this study, we investigated impact chronic on its metabolic profiles mice. Our results revealed that proportion Firmicutes Bacteroidetes was not altered, while abundance Proteobacteria Actinobacteria significantly decreased. addition, content lipids abundant increased. A pathway analysis differential metabolites suggested numerous pathways were affected, glycerophospholipid metabolism most altered. Combined variations function intestinal microorganisms closely relevant Actinobacteria. conclusion, long-term diversity composition microbiota mice, substantially altered microbiome. This may represent novel mechanism which an unhealthy lifestyle such as mistimed coffee breaks lead exacerbates disease.

Abstract Background: Incidence of early-onset colorectal cancer (EOCRC; e.g., diagnosed before age 50) in the United States has increased substantially since 1990s but underlying reasons remain unclear. Methods: We examined ecologic associations between dietary factors and EOCRC incidence adults aged 25–49 during 1977–2016 States, using negative binomial regression models, accounting for age, period, race. The models also incorporated an age-mean centering (AMC) approach to address potential confounding by age. stratified analysis sex computed rate ratio (IRR) each study factor. Study factor data (for 18 variables) came from repeated national surveys; Surveillance Epidemiology, End Results Program. Results: suggest that on association with likely existed certain (e.g., calcium intake), AMC can alleviate confounding. was positively associated smoking [IRR (95% confidence interval (CI): 1.17 (1.10–1.24) men; 1.15 (1.09–1.21) women] alcohol consumption CI), 1.08 (1.04–1.12) (1.04–1.11) women]. Claudin-2 (CLDN2), a component of tight junctions, is abnormally expressed in human lung adenocarcinoma tissue. CLDN2 contributes to chemoresistance adenocarcinoma-derived A549 cells, and it may be target for cancer therapy. Here, we found that coffee ingredients, namely caffeine theobromine, decreased the protein level cells. In contrast, other components, such as theophylline chlorogenic acid, had no effect. These results indicate 7-methyl group methylxanthines play key role reduction expression. The caffeine-induced was inhibited by chloroquine, lysosome inhibitor. protein-stability assay using cycloheximide, levels faster caffeine-treated cells than vehicle-treated suggest accelerates lysosomal degradation CLDN2. accumulation cytotoxicity doxorubicin were dose-dependently increased, which exaggerated but not spheroids. Caffeine nuclear factor-erythroid 2-related factor 2 (Nrf2) without affecting hypoxia-inducible factor-1α levels. Furthermore, expression Nrf2-targeted genes. effects on anticancer-drug-induced toxicity also observed RERF-LC-MS We enhances doxorubicin-induced spheroids mediated Nrf2

Rationale Nasopharyngeal carcinoma (NPC) is a malignant tumor that endemic in Southeast Asia, North Africa, and southern China. There an urgent need for effective early diagnosis treatment of this disease since NPC currently often detected at advanced stages. Methods To reveal the underlying metabolic mechanisms discover potential diagnostic biomarkers NPC, we employed ultrahigh‐performance liquid chromatography coupled with quadrupole time‐of‐flight mass spectrometry (UHPLC‐Q‐TOF‐MS) UHPLC‐Q‐Exactive Orbitrap MS, respectively, to analyze 54 serum samples urine from 27 patients healthy control individuals. Results A total 1230 metabolites were determined samples, 181 significantly changed patients. The enriched 16 pathways, including biosynthesis unsaturated fatty acids, cholesterol metabolism, ferroptosis. 2509 samples. Among them, 179 altered patients, these eight tricarboxylic acid (TCA) cycle caffeine metabolism. Seven metabolites, creatinine paraxanthine, found be both Based on further biomarker analysis revealed panel three octanoylcarnitine, creatinine, decanoyl‐l‐carnitine, displayed perfect performance (area under curve [AUC] = 0.973) distinguish controls, while other three‐metabolite panel, consisting stachydrine, had AUC 0.809 Conclusion This work highlights key pathways disturbed presents disease.
