A growing body of research suggests that locus coeruleus-norepinephrine (LC-NE) system may function differently in individuals with autism spectrum disorder (ASD). Understanding the dynamics both tonic (resting pupil diameter) and phasic (pupil dilation response [PDR] event-related potential [ERP]) indices provide meaningful insights about nature LC-NE ASD. Twenty-four children ASD 27 age- nonverbal-IQ matched typically developing (TD) completed two experiments: (1) a resting eye-tracking task to measure diameter, (2) three-stimulus oddball paradigm responsivity using PDR ERP. Consistent prior reports, our results indicate exhibit increased reduced (PDR ERP) activity compared their TD peers. For groups, decreased was associated diameter. Lastly, were primarily related measures attention-deficit/hyperactivity (ADHD), not ASD, symptomatology. Detection of deviant stimuli is crucial to orient and adapt our behavior. Previous work shows that elicit phasic activation the locus coeruleus (LC), which releases noradrenaline controls central arousal. However, it unclear whether detection behaviorally relevant selectively triggers LC responses or other neuromodulatory systems (dopamine, serotonin, acetylcholine). We combine human functional MRI (fMRI) recordings optimized for brainstem imaging with pupillometry perform a mapping deviant-related in subcortical structures. Participants have detect items "local-global" paradigm distinguishes between deviance based on stimulus probability sequence structure. fMRI are distributed many cortical areas. Both types pupil, LC, systems. Our results reveal task-relevant recruits same multiple across computationally different deviance.

Pupil size variations have been associated with changes in brain activity patterns related specific cognitive factors, such as arousal, attention, and mental effort. The locus coeruleus (LC), a key hub the noradrenergic system of brain, is considered to be regulator control on pupil size, diameter corresponding release norepinephrine (NE). Advances eye-tracking technology open-source software facilitated accurate measurement various experimental settings, leading increased interest using pupillometry track nervous activation state potential biomarker for disorders. This review explores non-invasive fully translational tool studying cortical plasticity starting from recent literature suggesting that could promising technique estimating degree residual human subjects. We investigated relationship between pupil diameter blink rate volume somatosensory cortex male female unanesthetized, head-fixed mice. monitored these variables while mice were awake, during periods of rapid eye movement (REM), non-rapid (NREM) sleep.
