Among sixty-eight pseudomonads, isolate QCS59 from the rhizosphere of H. schimperi was selected based on its siderophore level. Production optimal in Kings B supplemented with 2% peptone and 0.5% fructose at pH 6.5 25 °C for 72 h. Additionally, threshold potential iron found a concentration 10 µM. After purification, acidified presented maximum absorption peak 360 nm, while neutral form 414 confirming pyoverdine (PVD) nature. Furthermore, major appeared retention time (RT) 27.5 min during RP-HPLC, homogeneity. Interestingly, it demonstrated effective antibacterial activity, especially against Escherichia coli ATCC 8739, minimum inhibitory (MIC) 6.3 µg/mL bactericidal (MBC) 12.5 µg/mL. At ½ MIC value, inhibited 82.1% well-established biofilms Salmonella enterica. There an increase malondialdehyde (MDA) antioxidative enzymes, catalase (CAT) treated bacteria because peroxidation membrane lipids oxidative stress, respectively. SEM proved cellular lysis surface malformation most bacteria. This study concludes that is promising candidate treating wastewater skin pathogens.

Bacterial social interactions play crucial roles in various ecological, medical, and biotechnological contexts. However, predicting these from genome sequences is notoriously difficult. Here, we developed bioinformatic tools to predict whether secreted iron-scavenging siderophores stimulate or inhibit the growth of community members. Siderophores are chemically diverse can be stimulatory inhibitory depending on bacteria have lack corresponding uptake receptors. We focused 1928 representative Pseudomonas genomes an experimentally validated coevolution algorithm match encoded siderophore synthetases receptor groups. derived community-level iron interaction networks show that siderophore-mediated differ across habitats lifestyles. Specifically, dense sharing competition were observed among environmental nonpathogenic species, while small, fragmented occurred human-associated pathogenic species. Together, our sequence-to-ecology approach empowers analyses thousands bacterial strains offers opportunities for targeted intervention microbial communities.

The global rise of antibiotic resistance calls for new drugs against bacterial pathogens. A common approach is to search natural compounds deployed by microbes inhibit competitors. Here, we show that the iron-chelating pyoverdines, siderophores produced environmental Pseudomonas spp., have strong antibacterial properties inducing iron starvation and growth arrest in screen 320 isolates used 12 human pathogens uncovered several pyoverdines with particularly high distinct chemical characteristics. most potent pyoverdine effectively reduced Acinetobacter baumannii , Klebsiella pneumoniae, Staphylococcus aureus a concentration- iron-dependent manner. Pyoverdine increased survival infected Galleria mellonella host larvae showed low toxicity host, mammalian cell lines, erythrocytes. Furthermore, experimental evolution combined whole-genome sequencing revealed limited compared an antibiotic. Thus, from strains potential become class sustainable antibacterials specific

Background/Objective:Staphylococcus aureus produces staphyloferrin A (Sfna) siderophores to sequester host iron during infection and rapid cell proliferation We examined drug susceptibility, siderophore production, genome sequencing of clinical isolates S. aureus. Methods: total 100 specimens, including pus, sputum, hemoculture, urine, tissue, fluid, skin scrap were grown in iron-deprived Luria broth agar. The investigated for spectral signature using MALDI–TOF/MS, while antibiotic susceptibility content assessed the chrome azurol S method. Whole partial 16S rRNA DNA sequences employed, VITEK/MS revealed specific spectra. Results: Clindamycin, erythromycin, gentamicin, linezolid, moxifloxacin, oxacillin, trimethoprim/sulfamethoxazole, vancomycin (100%) most common antibiotics which susceptible. Sfna was not detectable fluid isolates, encoded by sfnaB, sfnaD, sfnaB/sfnaD genes. However, they pus (73.8%), sputum (85.3%), hemoculture (50.0%), urine (85.7%) isolates. subspecies, JKD6159, SA268, MN8, found be 72.73% according sequencing. Conclusion: staphylococci sensitive detected resulting production sfna

One Health is an integrative concept striving for healthy environments, animals, and humans. Microbes play a fundamental role this as they are key drivers of ecosystem stability host health. A central microbial feature across various environments the secretion siderophores, low-molecular-weight metabolites that chelate ferric iron (Fe3+) with high affinity. We propose that, by steering metal availability, siderophores can be important in four areas via: (i) enhancing crop yield, (ii) mitigating heavy pollution, (iii) preventing pathogen colonization, (iv) serving potential treatments established infections. Collectively, attractive molecules to complement our efforts increasing human, animal, environmental
